小鼠移植性S180肉瘤模型的建立及温和加热对热休克蛋白70表达的影响

摘    要:
目的 :研究NH小鼠S180肉瘤细胞移植后肿瘤模型生长特点 ,以及热休克蛋白 (heatshockproteinsHSPs)70在温和加热 (4 2℃ )不同时段表达变化。方法 :不同剂量S180肉瘤细胞于NH小鼠背部皮下接种 ,观察肿瘤组织生长规律 ,取部分肉瘤细胞进行热休克处理 2、 4、 6、 8、 10、 12h后 ,进行HSP 70免疫组化观察 ,结果 :肿瘤发生率10 0 % ,不同剂量组的肿瘤生存期限差异有显著意义 ,肿瘤生长大小无显著差异 ;对HSPs的免疫组化观察表明 :热休克处理后HSP 70阳性增强 ,以 6h后最明显 ;8h后细胞发生皱缩、溶解。结论 :此模型是HSPs/肽复合物提取、纯化中稳定的动物肿瘤疫苗治疗模型 ;分离、纯化HSP 70时将肿瘤组织热休克处理 (4 2℃ ) 6h即可。

Abstract:
Objective: To study the development characteristics of S180 sarcoma model in NH mice and the expression rule of HSP 70 in different time after heat shock.Method:The rule of tumor growth was observed after the S180 sarcoma cells were inoculated at NH mice back subcutaneous in different dose and immunohistochemical expression of HSP 70 was observed.Result:Sarcoma were formed in 100%.There were significant difference between vary dose in survival time,the HSP 70 expression was enhanced after heat shock treated especially at 6 hours.The sarcoma cells crimpled and dissolved after 8 hours.Conclusion:The S180 sarcoma model in mice is a good model to purify HSPs/peptide complexes in tumor vaccine therapy.Heat shock treatment of 6 hours is suitable to purify HSP 70.
