中药白芨建立大鼠脑缺血模型可行性探讨

摘    要:目的:以中药白芨作为栓塞剂建立大鼠脑缺血模型,探讨这一新方法的可行性. 方法:手术暴露大鼠右侧颈部血管,向右颈内动脉注入一定量的白芨微粒悬液或白芨胶,在术后一定时间内进行神经功能缺失评分、大脑切片红四氮唑(TTC)染色和组织学H.E染色. 结果:白芨微粒组大鼠术后4 h内呈现意识障碍,持续性对侧肢体轻度偏瘫及环转,TTC染色显示右侧大脑半球额叶、顶叶皮层和基底核附近局灶性缺血或梗塞,光学显微镜下可见缺血区脑组织水肿或坏死,神经元轮廓模糊,核固缩或裂解等;白芨胶组仅在术后术后1 h内有轻度意识障碍,12 h内出现对侧肢体行动障碍,但未见明显病理改变. 结论:白芨微粒可以作为栓塞剂建立大鼠脑缺血模型.

Abstract:AIM: To develop a new embolic model of cerebral ischemia induced by common bletilla tuber in Wister rats. METHODS: The suspension of bletilla particles or bletilla gum was injected into the right internal carotid artery after the external carotid artery was exposed in Wister rats. In a certain period after operation, the assessment of neurological defects, red tetrazoline (TTC) staining and HE staining were performed. RESULTS: Four hours after operation, rats injected with bletilla particles developed temporary conscious disturbance, and sustained hemiparesis of contralateral limbs with spontaneous circling behavior. Ischemia or infarction was found obviously in the cortex of parietal, frontal lobes and basal nuclei of the right cerebral hemisphere, where the neurons characterized with vague outlines, pyknosis and necrosis of neuron nuclei were recognized as well. But hemiparesis and spontaneous circling behavior only lasted for 12 h after embolization with bletilla gum. CONCLUSION: Bletilla particles can be used to develop a sustained cerebral ischemia model in rats.
