人膀胱癌耐药细胞系BIU-87/A、BIU-87/V的建立及耐药分析

摘    要:为研究人膀胱癌细胞株耐药机制,应用阿霉素(ADM)、足叶乙甙(VP-16)低浓度持续递增法,在体外持续培养10个月,逐步诱导人膀胱癌细胞系BIU-87,获得具有多重抗药性的BIU-87/A、BIU-87/V细胞。通过检测抗癌药物50%抑制率剂量,BIU-87/A、BIU-87/V细胞分别对ADM、VP-16的耐药程度较亲本细胞提高57.42倍、5.44倍。两种耐药株显示了相似的耐药谱,均对ADM、VP-16、长春新碱和阿糖胞苷等药物产生耐药性,且随着培养时间延长,耐药性增强,但对丝裂霉素C、噻替哌和顺铂仍保持敏感性。结果表明BIU-87/A、BIU-87/V细胞具有多重抗药性(MDR)表型,为不同药物诱导人膀胱移行细胞癌MDR机制和筛选抗药性逆转剂提供理想模型。

Abstract:To study the resistance mechanism of human bladder cancer cell line, an adriamycin(ADM) resistance bladder cancer cell line, BIU 87/A and an etoposide (VP 16) resistance cell line, BIU 87/V were established by exposing the BIU 87 parent cells to a culture medium with a persistent increase of concentration of ADM or VP 16 respectively over a period of 10 months. The BIU 87/Acells became resistance to ADM 57.42 times , and the BIU 87/V cells to VP 16 5.44 times stronger than their parent cells at the concentration that caused 50 % inhibition of cell growth and exhibited cross resistance to ADM,VP 16,vincristine and alexan but not to mitomycin C, thiotepum and cisplatin. When BIU 87/A and BIU 87/V were cultured in the increasing dose drug continuously, the drug resistance property was more stronger. It was suggested that BIU 87/A and BIU 87/V had the phenotype of multidrug resistance with relative stability and could serve as an ideal model for the study of multidrug resistance in human bladder transitional cell carcinoma.
