摘    要:
目的探索快速检测21号染色体的方法.方法对8例外周血淋巴细胞培养标本分别进行引物原位标记反应和常规细胞遗传学分析.结果中期分裂相21号染色体标记率平均为98%(97%~100%);平均91%(89%~95%)的间期细胞核显示2个信号.荧光信号清晰明亮,引物原位标记反应在4~5 h内完成.其结果与常规G显带结果一致.结论引物原位标记技术检测外周血标本中21号染色体特异性强,标记效率高,可作为产前快速诊断异倍体的新方法.

Abstract:
Objective To detect human chromosome 21 rapidly. Methods The Primed in situ labeling (PRINS) reactions were performed on 8 preripheral lymphocyte sample with chromosome 21 specific oligonucleotide primer. Results Primer 21 provided strong and well defined signals on both metaphases and interphase nuclei samples. The efficiency of labeling was 97 %-100 % (mean = 98 %) and 89 %-95 % (mean = 91 %-) in metaphases and interphase nuclei respectively. Conclusion PRINS may be a raliable technique for prenatal detecting Chromosome 21 aneuploidies.
