12-脂氧化酶影响胃癌细胞株AGS蛋白激酶C活性

摘    要:
目的 研究胃癌细胞中蛋白激酶C(PKC)活化与12-脂氧化酶(12-LOX)的关系.方法 胃癌细胞株AGS于含10%胎牛血清的RPMI 1640培养液中常规培养,AGS细胞培养24 h至对数生长期改用无血清RPMI 1640培养基培养24 h加入干预药物.PKC活性采用非同位素标记法测定.分别采用12-LOX催化合成产物12-羟基廿碳四烯酸(12-HETE)及12-LOX特异性抑制剂--黄芩素干预AGS细胞,测定其PKC活性变化.结果 AGS细胞经12-HETE干预后PKC活性明显增高,而经黄芩素干预后PKC活性明显下降,且呈时间、剂量依赖性.结论 12-LOX对PKC活性具有调节作用;PKC参与了12-LOX影响胃癌细胞增殖凋亡的信号传导.

Abstract:
Objective To investigate the effects of 12-lipoxygenase(12-LOX) and its inhibitor, baicalein, on PKC activation in human gastric cancer cell AGS. Methods Human gastric cancer cell AGS were cultured in a RPMI 1640 medium with 10% fetal bovine serum(FBS). After overnight incubation, AGS cells were cultured in a fresh RPMI 1640 medium without FBS for another 24 h and then treated with 12-LOX inhibitor, baicalein, or its metabolite 12-HETE respectively in various concentrations. The activity of PKC was examined by PePTag non-radioactive protein kinase assay. Results The PKC activity was increased after 12-HETE cocultured(P<0.05) and decreased after baicalein(P<0.05) in a time and concentration dependent manner. Conclusion 12-LOX affects the activity of PKC in gastric cancer cell line AGS. PKC is involved in the proliferation or apoptosis procedure mediated by 12-LOX in AGS cells. 12-LOX inhibitor may contributed an anticancer affect in human gastric carcinoma.
