依达拉奉对脑出血大鼠细胞凋亡及Bcl-2、Bax蛋白表达的影响

摘    要:
目的研究依达拉奉对脑出血大鼠血肿周围脑组织细胞凋亡及Bcl-2、Bax蛋白表达的影响,探讨其对脑出血后脑组织损害的保护作用。方法36只SD大鼠随机分为依达拉奉组、脑出血组和假手术组,每组12只。用尾状核注射自体血法建立脑出血模型;检测丙二醛(MDA)含量,以TUNEL法及免疫组化法检测神经细胞凋亡数及Bcl-2、Bax蛋白表达的变化。结果出血组脑组织MDA含量、细胞凋亡数和Bcl-2、Bax蛋白表达水平均明显高于假手术组;与出血组相比,依达拉奉组MDA含量、细胞凋亡数与Bax蛋白表达水平均下降,而Bcl-2蛋白表达水平显著增高。结论依达拉奉可抑制出血后神经细胞凋亡,此作用可能与其减轻氧化应激、上调Bcl-2和下调Bax蛋白表达水平有关。

Abstract:
Objective To investigate the effect of edaravone on the cell apoptosis and the expression of Bcl-2 and Bax protein following intracerebral hemorrhage(ICH) in rats,and explore the protective effect of edaravone on the cerebral injury induced by hemorrhage.Methods Thirty SD rats were randomly divided into 3 groups:edaravone-treated group,ICH group and sham group(n=12).The rat model of ICH was established by injection of autologous blood into the caudate nucleus.The content of malondialdehyde(MDA) in perihematoma was detected by spectrophotometry.Neuronal apoptosis and the expression of Bcl-2 and Bax protein were monitored by TUNEL and immunohistochemistry,respectively.Results The content of MDA,apoptosis and the expression of Bcl-2 and Bax protein were significantly increased in the ICH group as compared with the sham group.Treatment of edaravone markedly reduced the content of MDA,apoptosis and the expression of Bax protein in comparison to the ICH group,while the expression of Bcl-2 was obviously higher in the edaravone-treated group than that in the ICH group.Conclusion Edaravone inhibits the neuronal apoptosis following intracerebral hemorrhage,which might be relation with the alleviated oxidative stress,up-regulated Bcl-2 protein and down-regulated expression of Bax protein.
