miRNA-671上调抑制胃癌细胞增殖与迁移

目的 探索miR-671在胃癌组织和细胞中的表达,并研究其对胃癌细胞增殖和迁移的影响.方法 通过qRT-PCR实验检测miR-671在胃癌组织和细胞中的表达,通过CCK-8实验检测miR-671表达上调对MKN45胃癌细胞增殖,通过划痕和Transwell小室实验检测miR-671表达上调对MKN45胃癌细胞侵袭和迁移的影响.结果 与癌旁正常组织相比,胃癌组织中的miR-671表达明显下调(P＜0.01);与正常人胃黏膜上皮细胞相比,胃癌细胞中的miR-671表达也明显下调(P＜0.01);miR-671表达上调明显抑制MKN45胃癌细胞增殖、迁移和侵袭(P＜0.01).结论 miR-671可以抑制胃癌细胞的增殖、迁移和侵袭,在胃癌中发挥抑癌基因功能.

Up-regulation of miRNA-671 inhibits proliferation and invasion of gastric cancer cells

Objective To investigate the expression of miR-671 in gastric cancer tissues and cell lines,and determine its effects on the proliferation and invasion of gastric cancer cells.Methods The expression of miR-671 was detected in gastric cancer tissues and para-carcerous tissues from 8 patients and cell lines SGC7901,MKN28 and MKN45,and human gastric mucosal epithelial cell line GES-1 by quantitative real-time PCR (qRT-PCR).CCK-8 assay was used to measure the cell proliferation after miR-671 upregulation.The effect of miR-671 on cell migration and invasion was detected by wound healing assay and Transwell chamber assay.Results The expression level of miR-671 was significantly decreased in gastric cancer tissues than the adjacent normal tissues (P ＜ 0.01).The expression levels of miR-671 were significantly decreased in the gastric cancer cell lines compared with GES-1 cells (P ＜0.01).Up-regulation of miR-671 significantly inhibited the proliferation,migration and invasion in gastric cancer cells (P ＜ 0.01).Conclusion miR-671 inhibits proliferation,migration and invasion of gastric cancer cells,and may serve as a tumor suppressor.
