电磁辐射致Raji细胞内Caspase-3、8蛋白表达的变化及其意义

摘    要:目的对比性研究电磁脉冲(electromagnetic pulse,EMP)、S波段高功率微波(S-band high power micro-wave,S-HPM)、X波段高功率微波(X-band high power microwave,X-HPM)三种不同波段电磁辐射对Raji细胞内Caspase-3、Caspase-8蛋白表达的影响,探讨其在电磁辐射生物学效应的相关调控机制。方法常规培养Raji细胞,用S-HPM、X-HPM和EMP三种不同波段的电磁辐射照射Raji细胞,设伪照射为对照组。于照射6 h收集细胞,提取总蛋白,采用Western blot技术检测照射后细胞内Caspase-3、Caspase-8两种蛋白的表达;应用图像分析系统对阳性条带进行定量分析,所得数据用SPSS13.0软件的一元方差分析进行统计学处理。结果三种不同波段的电磁辐射照射后6 h后均可导致Raji细胞Caspase-3、Caspase-8蛋白表达发生改变,与对照组相比较,各实验组Raji细胞Caspase-3蛋白表达均明显上调(P〈0.01),EMP与X-HPM组间无统计学差异(P〉0.05),但均较S-HPM组明显上调(P〈0.01),且表达程度呈X-HPM〉EMP〉S-HPM的趋势;Caspase-8只在X-HPM照后明显上调(P〈0.01),但在EMP、S-HPM照射后均明显下调(P〈0.01);EMP较S-HPM下调更明显(P〈0.01)。结论三种电磁辐射均可导致Raji细胞Caspase-3、Caspase-8蛋白表达的改变,其中X-HPM主要通过Caspase-8进一步诱导并激活Caspase-3发挥作用,而EMP和S-HPM可能通过其他途径激活Caspase-3而发挥作用。

Abstract:Objective To compare the effects on Caspase-3 and Caspase-8 protein expressions in Raji cells induced by electromagnetic pulse(EMP),S-band high power microwave(S-HPM) and X-band high power microwave(X-HPM),and investigate the molecular mechanism.Methods Raji cell line was regularly cultured and radiated by EMP,S-HPM and X-HPM respectively.The control group received pseudo radiation.Cells were collected at the sixth hour after radiation.Western blot technique was performed to examine the expressions of Caspase-3,8 proteins respectively.The positive signal bands of Caspase-3,8 were quantified with image analysis system.The obtained data were analyzed by SPSS13.0 software.Results The expressions of Caspase-3,8 proteins were changed after electromagnetic radiation.Caspase-3 protein was significantly upregulated after EMP,S-HPM and X-HPM radiation(all P<0.01),and showed a regularity as X-HPM >EMP>S-HPM.Caspase-8 was significantly upregulated after X-HPM radiation(P<0.01),and downregulated after S-HPM and EMP radiation(both P<0.01).Conclusion The three different bands of electromagnetic radiation can induce changes of Caspase-3,8 protein expressions.X-HPM might active Caspase-3 through Caspase-8 pathway,while EMP and S-HPM through other pathways.
