应用噬菌体随机肽库筛选腺病毒模拟肽的研究

摘    要:目的 探讨模拟腺病毒表位肽的噬菌体对该病毒感染Hela细胞的保护作用,以便为研制防治腺病毒感染的新型疫苗奠定基础。方法 用3型腺病毒中和抗体筛选噬菌体随机六肽库,经3轮筛选后,随机挑取了21个噬菌体克隆,扩增后分别加入3型腺病毒感染的Hela细胞,用结晶紫染色法观察Hela细胞的病变程度。结果 10/21个克隆噬菌体短肽对3型腺病毒感染Hela细胞具有保护作用。结论 噬菌体表面的短肽可以模拟中和抗体相关的腺病毒表位,对3型腺病毒感染Hela细胞有一定的保护作用。

Abstract:Objective To investigate the protective effects of minotopes of adenovirus on Hela cells infected with the adenovirus,in order to lay a foundation for developing the vaccine to prevent and treat adenovirus infection. Methods Type 3 adenovirus neutralizing antibody was used to screen phage displayed 6 mer peptide library.After 3 rounds of biopanning,21 phage clones were selected randomly and added to the Hela cells infected with adenovirus. The pathological changes of the cells were observed by crystal violet staining. Results Of the 21 phages clones,10 clones showed protective effects on the Hela cells infected with type 3 adenovirus respectively. Conclusions Random phage mimotopes recognized by type 3 adenovirus neutralizing antibody could protect Hela cells from being infected with type 3 adenovirus.
