bFGF对高眼压下兔视网膜神经节细胞的保护作用

摘    要:目的:研究碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)对高眼压下兔视网膜神经节细胞的保护作用。方法:每周1次前房注入甲基纤维素制成慢性高眼压动物模型,随机将动物分为高眼压对照组、外科对照组和bFGF治疗组,另加1组正常对照组。治疗组每周1次玻璃体内注射bFGF,高眼压对照组不作任何处理,外科对照组仅于玻璃体内注入与治疗组等量的磷酸缓冲生理盐水(plosphate buffered saline,PBS),实验持续4wk。每周测定各组动物的图像视网膜电图(pattern electroretinogram,PERG),观察各组动物b波变化趋势,最后光镜、电镜观察动物视网膜神经节细胞的超微结构变化。结果:治疗组PERGb波振幅仅见轻微下降趋势,与正常组接近,两对照组则下降趋势显著,结果经方差分析有统计学意义;形态学上可见治疗组节细胞超微结构变化轻微,对照组则变化明显。结论:bFGF作为一种神经营养因子,对高眼压下兔视网膜神经节细胞具有保护作用。

Abstract:Objective: To explore the protective role of basic fibroblast growth factor (bFGF) to rabbit's retinal ganglion cells under high intraocular pressure. Methods: Rabbits with continuous elevated intraocular pressure induced by injection of methyl cellulose into anterior chamber once a week were divided into three groups at random: high intraocular pressure group, surgical control group and treatment group, a normal group was set for comparing. bFGF was injected into vitreous body in treatment group once a week, there was no treatment to high intraocular pressure group, an equal volume of phosphate buffered saline was injected into vitreous body in surgical control group as well as treatment group, the experiment lasted for 4 weeks. Pattern electroretinogram (PERG) was tested in all groups once a week, and the b-wave amplitude curve was studied in all groups. Finally, ultrastructure changes of retinal ganglion cells were examed through light and electron microscope. Results: The b-wave amplitude curve in treatment group descended slightly, similar to normal group, while it descended distinctly in two control groups, analysis of variance was used as statistic method. Only slight ultrastructure changes in treatment group could be seen comparing to the control groups. Conclusion: As a kind of neurotrophin, bFGF can protect rabbit's retinal ganglion cells under high intraocular pressure.
