埃克特注射液体内外抗肝癌作用研究

摘    要:目的:观察埃克特注射液对人肝癌细胞SMMC-7721细胞及小鼠移植性肿瘤H22的抑制作用。方法:用MTT法、细胞生长曲线实验研究不同浓度的埃克特注射液作用48h对SMMC-7721细胞的生长抑制作用,Hoechst 33258染色观察细胞凋亡,以了解该药物对肝癌细胞的体外增殖作用;同时建立小鼠H22肝癌移植性实体瘤模型研究埃克特注射液对肝癌的体内抑制作用。结果:埃克特注射液对SMMC-7721具有抑制作用,呈明显的量效关系,IC50为2.325mg/L;生长曲线提示埃克特注射液对SMMC-7721细胞的抑制作用呈明显时效和量效关系;Hoechst 33258染色后,给药组出现明显的细胞凋亡形态学变化;腹腔注射0.5、1、2mg/kg埃克特注射液对小鼠移植性肿瘤H22的抑制率分别为34.37%、42.36%、57.99%,呈剂量依赖性。结论:埃克特注射液对人肝癌细胞SMMC-7721细胞具有明显体外抗增殖作用,对小鼠移植性肿瘤H22有一定的抑制作用。

Abstract:Objective: To investigate the inhibitory effect of Aikete injection on human hepatic cancer cell line SMMC-7721 and hepatic cancer H22 . Methods : MTT assay and growth curve assay were used to assess the inhibitory effect of Aikete injection on proliferation of SMMC-7721 cells after treated for 48 hours.Hoechst 33258 staining was used to observe cell apoptosis .Hepatic cancer H22 model in mice was performed to study anti-hepatocarcinoma activity of Aikete injection in vivo. Results: After treated with Aikete injection for 48 hours, MTT assay showed Aikete injection can inhibit the proliferation of SMMC-7721 cells and have obvious dose-effect relationship. IC50 of Aikete injection to SMMC-7721 cells was 2.325mg/L. The dose-effect relationship and time-effect relationship were described in growth curve. Nuclear pyknosis and chromatin condensation which were the characteristics of cell apoptosis were observed by Hoechst 33258 staining. The experiment in vivo showed Aikete injection of 0.5,1,2 mg/kg ip had significant inhibitory effect on tumor and dose-related effect in mice with hepatocarcinoma. Conclusions: Aikete injection could inhibit proliferation of SMMC-7721 cells in vitro and the growth of hepatic cancer H22 in vivo.
