粉葛的高效液相色谱指纹图谱

摘    要:目的对不同来源的葛根药材指纹图谱进行比较,为葛根的质量控制提供可靠方法。方法应用RP HPLC法。色谱柱为Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇水,流速0.5 mL.min-1,检测波长250 nm,柱温30℃,以葛根素为参照物。结果通过对11批不同产地粉葛药材的测定,初步建立了粉葛的HPLC指纹图谱,标定了10个共有峰。通过中药指纹图谱计算机辅助相似度软件计算出其相似度均在0.9以上。结论RP HPLC法能较好地识别葛根药材,为葛根的质量控制提供方法学依据。

Abstract:Objective To make comparative study on HPLC-FPS of several kinds of Pueraria lobata and P.thomsonii from different sources,and provide some useful information for quality evaluation.Methods A Kromasil C_(18) column(250 mm×4.6 mm,5 μm) was used with a solvent system consisting of methanol and water in a gradient mode with a flow rate of 0.5 mL·min~(-1),and the detecting wavelength was set at 250 nm.The column temperature was maintained at 30 ℃.Results The fingerprint of Radix Puerariae was established and 10 common peaks were pinpointed by the determination of the 11 P.thomsonii Benth.samples gathered from different places.The similarities,which were proved to be higher than 0.9,were calculated with the help of the similarity calculation soft of fingerprint chromatography of traditional Chinese medicine.Conclusions The method and data can be used to control the quality of Radix Puerariae.
