大鼠肌源性干细胞分化为神经样细胞过程中Hes1蛋白的表达差异

摘    要:目的:研究Hes1蛋白在大鼠肌源性干细胞(MDSCs)体外诱导分化为神经样细胞过程中的表达差异。方法:大鼠乳鼠骨骼肌消化后体外培养增殖,第5代传代后细胞分为两组:对照组完全培养基培养,实验组加入诱导剂培养。6d后观察两组细胞的形态变化,并进行神经元特异性烯醇化酶(neuron-specific enolase,NSE)免疫细胞化学鉴定。然后通过免疫荧光和Western Blot技术观察Hes1蛋白的表达差异。结果:实验组细胞呈NSE染色阳性,对照组基本不表达;免疫荧光和Western Blot结果显示,与对照组比较,实验组Hes1蛋白表达较强(P0.05)。结论:在体外,大鼠肌源性干细胞分化为神经样细胞过程中Hes1蛋白表达降低。

Abstract:Objective:To investigate the differential expression of Hes1 protein during the proliferation and differentiation of muscle derived stem cells(MDSCs) into neuron-like cells in vitro.Methods:The muscle-derived stem cells from the neonate rat were cultured and purified clonally in vitro.The 5 th passage cells were divided into 2 groups.The control group was not intervened,only cultured by complete medium;and the experiment group was added with inductor.Morphological changes and neuron-specific enolase(NSE) immunocytochemistry were observed after six days.Immunofluorescence and Wescern Blot technigues were used to demonstrate the Hesl expressional difference.Results:The cells in the experiment group were observed NSE-immunoreactivities,but the control group was negative.Immunofluorescence and Western Blot showed that the expression level of Hes1 protein in the experiment group was much higher than that of the control group(P<0.05).Conclusion:In vitro,the expression of Hes1 protein is decreased when the MDSCs were induced into neuron-like cells.
