P38MAPK在妊娠期糖代谢异常大鼠肾脏组织中的表达

摘    要:
目的初步研究P38MAPK在妊娠期糖代谢异常大鼠肾脏组织中的表达水平。方法选取雌性sD大鼠60只,雄性SD大鼠15只,分为高糖高脂饮食-孕鼠组(SFP)和非孕鼠组(SFV),普通饮食一孕鼠组(NP)和非孕鼠组(NV)。相应组分别受孕,于妊娠第20天四组大鼠心内取血,检测血糖、胰岛素水平,并计算胰岛素抵抗指数。用RT—PCR法检测大鼠肾脏组织中P38MAPK的表达水平。结果SFP组的空腹血糖值、胰岛素水平、胰岛素抵抗指数与其他三纽相比,差异均有统计学意义(P〈0.05),证明妊娠期糖代谢异常大鼠模型建立成功;P38MAPK mRNA在肾脏组织中有表达,并且:①P38MAPKmRNA在SFP组的表达水平明显升高,与其他三组相比差异有统计学意义(P〈0.01)。②P38MAPKmRNA在SFV组和NP组的表达水平均升高,但是二者之间差异无统计学意义(P〉0.05)。⑧P38MAPK mRNA在SFV组和NP组的表达水平与NV组相比升高明显,差异有统计学意义(P〈0.05)。结论P38MAPK可能参与了妊娠期糖代谢异常疾病的肾脏损伤过程。

Abstract:
Objective To study the expressions of P38 mitogen -activated protein kinase (P38MAPK)in kidneys of pregnant rats with abnormal glucose metabolism. Methods Sixty female SD rats and fifteen male SD rats were divided into four groups: sucrose and fat diet pregnant rats (SFP), sucrose and fat diet virgin rats (SFV), normal diet pregnant rats (NP), normal diet virgin rats (NV). Mating with two fe- males and one male, the first day of gestation was estimated by the presence of spennatozoids in vaginals meats. On days 20 of gestation fasting blood glucose and insulin were assayed by acquiring from the rat heart, caeulate the insulin resistance index (IRI), RT - PCR was used to detected P38MAPK mRNA expression in kidney. Results In SPF group, the fasting blood glucose, insulin and IRI were significantly higher than that of other 3 groups (P 〈 0.05), proved that the model of pregnant rats with abnormal glucose metabolism has been established successfully. RT--PCR showed that:①The expression of P38MAPK mRNA in SPF group were significantly higher than that of other 3 groups ( P 〈 0. 01 ) ; ②There was no significant difference for the level of P38MAPK mRNA between SFV group and NP group( P 〉0.05) ; ③The level of P38MAPK mRNA of SFV group and NP group were higher than that of NV group ( P 〈 O. 05 ). Conclusions P38MAPK maybe involed in the kidney injury of diseases during gestational with glucose disorder.
