生物质谱技术及其在核酸领域的应用

摘    要:
生物质谱技术具有准确、快速、灵敏度高等特点,因此在生物大分子领域得到了广泛应用。本文介绍了电喷雾电离和基质辅助激光解吸/电离质谱技术的基本原理,并综述了生物质谱在核昔酸的鉴定与定量分析、单核苷酸多态性分型以及寡聚核苷酸片断序列测定中的应用。

Abstract:
Objective:The advantage of biological mass spectrometry is accuracy,high speed,high sensitivity,and it is widely applied in biomacromolecule domain.The principle of electrospray ionization-mass spectrometry (ESI-MS) and matrix assisted laser desorption/ionization-mass spectrometry (MALDI-MS) is described in this text.Simultane- ously,its application in single nucleotide polymorphisms,characterization and quantification of nucleotide,and se- quencing of oligonucleotide is reviewed.
