脑康饮对脑缺血模型大鼠学习记忆功能的影响

摘    要:为探讨脑康饮(由黄芪、葛根、川芎、蒲黄等组成)治疗血管性痴呆(VD)的疗效与机理。将75只雄性Wistar大鼠随机均分为假手术组(A)、模型组(B)、中药小剂量组(C)、中药大剂量组(D)、西药组(E)5组。除A组外,其余动物结扎双侧颈总动脉形成慢性低脑灌注模型。造模第3d起,后三组分别以脑康饮小、大剂量和脑复康灌胃治疗,期间以Y型迷宫进行学习记忆能力测试,50d后处死动物,测定脑组织胆碱能M受体(mACh-R)密度和乙酰胆碱酯酶(AchE)活性。结果:经中西药物治疗后,模型动物的学习记忆能力提高,脑mACh-R结合容量提高、AchE活性降低;且中药优于西药,中药大剂量优于小剂量。提示脑康饮可改善脑组织低灌流引起的学习记忆功能,这种作用可能与该药对脑内胆碱能系统的作用有关。

Abstract:To investigate the clinical effects and mechanism of Nao Kang Liquid in the treatment of vascular dementia (VD),75 male rats were randomized into group A,group B,group C,group D and group E. Except for group A, the rats in other groups were modeled into chronic low-flow perfusion by ligating their common carotid arteries. From the 3rd day after ligation,the rats in the latter three groups were treated with stomach perfusion of small dose of Nao Kang Liquid, large dose of Nao Kang Liquid and Nao Fu Kang respectively.During the treatment,the learning and memorizing abilities were tested by Y-shaped maze.Fifty days later,all rats were sacrificed and the density of mACh-R and the activity of AchE in the brain tissues were assayed.Results:After treatment,the learning and memorizing abilities improved;the content of mACh-R increased and the activity of AchE decreased,with better effects in groups C and D than in group E,group D than group C.It is revealed that Nao Kang Liquid can improve the learning and memorizing abilities in low-flow pefusion rats possibly by acting on cerebral cholinergic system.
