硫酸镍对大鼠睾丸细胞氧化应激效应的影响

摘    要:
目的观察硫酸镍(NiSO4)亚慢性染毒后对大鼠睾丸细胞的氧化作用。方法健康雄性Wistar大鼠32只,随机分为4组:生理盐水(NS)组,硫酸镍(NiSO4)1.0,2.0,4.0 mg/kg组。腹腔注射染毒,0.2 ml/100(g.bw),1次/d,连续30 d。制备睾丸组织匀浆,采用分光光度法检测睾丸组织中脂质过氧化物(LPO)、总抗氧化能力(T-AOC)、抗-活性氧(anti-ROS)、谷胱甘肽过氧化物酶(GSH-Px)水平的变化。结果染毒组与对照组比较,大鼠睾丸脏器系数无明显变化(P>0.05)。镍可诱导睾丸组织中LPO含量升高,2.0,4.0 mg/kg NiSO4组与对照组比较差异有统计学意义(P<0.05);2.0,4.0 mg/kg NiSO4组anti-ROS和GSH-Px活性低于对照组(P<0.01);各染毒组T-AOC均低于对照组(P<0.01)。结论镍对大鼠生殖系统的损伤可能与其所致的睾丸细胞氧化损伤有关。

Abstract:
Objective To study the oxidative stress effect of nickel sulfate(NiSO_4) on rats'testis,and to provide the laboratory theoretical evidence for preventing and curing of male reproductive toxicity.Methods 32 male Wistar rats with sexual maturation were divided into four groups randomly,and three groups of rats were administrted intraperitoneally with nickel sulfate daily at doses of 1.0,2.0,4.0?mg/kg respectively for 30 days,but the control group was administrated with saline in same volume according to the rats'weight.The testis homogenates were prepared to detect the levels of lipid peroxidation(LPO),total antioxide capacity(T-AOC),antireactive oxygen species(anti-ROS),glutathioe perioxidase(GSH-Px)in spectro photometric assay.Results The organ coefficient of testis exposured to NiSO_4 had no change compared with the control group(P>0.05).The LPO contents in testis of the 2.0,4.0?mg/kg groups were higher than that in the control group(P<0.05);The activities of anti-ROS and GSH-Px of the 2.0,4.0?mg/kg groups were lower than those in the control group(P<0.01).The T-AOC levels of the exposure groups were decreased compared with the control group(P<0.01).Conclusion The male reproductive toxicity is related to the enhancement of oxidative stress effect induced by nickel sulfate.
