还脑益聪方对早期认知功能障碍大鼠海马病理形态、炎症因子和氧化应激指标的影响

摘    要:
目的观察还脑益聪方对复合因素所致早期认知功能障碍大鼠行为学及海马病理形态、炎症因子和氧化应激指标的影响。方法采用皮下注射D-半乳糖及饲喂半高脂饲料的方法造成早期认知功能障碍大鼠模型,并以还脑益聪方灌胃治疗。采用Morris水迷宫试验观察大鼠行为学变化,HE染色及透射电镜观察海马病理形态学改变;应用放射免疫法检测大鼠海马匀浆白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量,酶联免疫法(ELISA法)测定氧化低密度脂蛋白(ox-LDL)含量,免疫比色法测定丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)和谷胱甘肽S转移酶(GSH-ST)含量。结果还脑益聪方能增加模型大鼠水迷宫试验穿台次数(P0.01),减轻模型大鼠海马神经细胞损伤、线粒体病变及胶质细胞增生,降低海马匀浆TNF-α及ox-LDL含量(P0.05,P0.01)。结论还脑益聪方能提高早期认知功能障碍模型大鼠的空间学习记忆能力,这可能与其抑制脑内炎症反应与氧化应激,从而减轻海马组织病理变化有关。
