兔椎间失稳软骨终板Ⅱ型胶原变化的相关实验研究

摘    要:目的:通过研究兔软骨终板Ⅱ型胶原在椎间失稳环境下的退变过程及机制,加深对椎间盘退变的认识。方法:选取48只6月龄日本大耳白兔雌雄不限,体重为(2.5±0.2)kg,均在相同条件下饲养,随机进行分组,分为正常对照组21只兔、实验失稳组27只兔;先将实验失稳组27只兔进行L6~7椎间手术失稳;均分别在术后2、4、6个月取材。对椎间盘软骨终板的Ⅱ型胶原用免疫组化的方法进行检测,用JD801图像分析系统进行图像分析,结果用SPSS11.5统计软件统计分析;以综合判断软骨终板的退变过程及机制。结果:(1)在自然增龄短期过程中,软骨终板的Ⅱ型胶原无明显变化;(2)椎间失稳可导致椎间软骨终板Ⅱ型胶原含量明显减少。结论:(1)在自然增龄过程中,软骨终板Ⅱ型胶原的无明显退变;(2)椎间失稳能明显导致椎间软骨终板Ⅱ型胶原的退变。

Degeneration of Rabbit Cartilage Endplate Collogen Ⅱ Due to Intervertebral Destabilization and Its Relevant Experimental Research

Abstract:Objective:By way of the study of the degeneration mechanism of the rabbit cartilage endplate in the circumstances of the intervertebral destabilization,we could find out the more cognition about the intervertebral discs degeneration. Methods:To choose forty-eight white macrotia rabbits of Japan, which weighed within the range of (2.5± 0.2) kg. female , male was unrestricted and were fed in the same conditions. These rabbits were divided into 2 groups randomly, with 21 rabbits in the normal contrast group, with 27 rabbits in the experimental destabilization group. First 27 rabbits of the experimental destabilization group were destabilizated by being operated on the L6-7. To get the materials from all groups after two months, four months and six months orderly. Collogen Ⅱ of the cartilage endplate were detected with the immunohistochemistry method, using JD801 picture analytical system to analyze it. Using SPSS11. 5 statistics soft ware to analyze it,then to judge the degenerative course and mechanism of the cartilage endplate synthetically. Results: (1)In the course of physio-aging,the cartilage endplate collogen Ⅱ diden't diversify obviously; (2)The intervertebral destabilization could cause the cartilage endplate to decrease the content of the collogen Ⅱ markedly. Conclusion: (1)In the course of physioaging the cartilage endplate collogen Ⅱ don't degenerate obviously; (2)The intervertebral destabilization can distinctly lead to the degeneration of the cartilage endplate collogen Ⅱ .
