摘    要:目的研究低氧时大鼠脑组织磷酸二酯酶(PDE)活性的变化及意义。方法18只SD大鼠随机分为正常对照组、窒息组和低氧-窒息组。低氧-窒息组6只予8%低氧气体吸入3h后窒息处死;窒息组6只予氮气窒息处死;正常对照组则用断头法处死。处死后迅速取脑制成脑组织酶样品,采用高效液相色谱法测定PDE活性。结果低氧-窒息组PDE活性为(230±28)nmol·g-1·min-1,明显低于正常对照组(365±67)nmol·g-1·min-1,而窒息组PDE活性(439±138)nmol·g-1·min-1则高于对照组,但差异无统计学意义。结论低氧时大鼠脑组织PDE活性可能存在一个时相的变化过程,PDE可能参与了缺氧性脑损伤的病理过程。

Abstract:Objective To study the changes and clinical significance of phosphodiesterase (PDE) activity in rat brain during hypoxia. Methods Eighteen Sprague-Dawley rats were divided into hypoxia-asphyxia group,asphyxia group and control group at random.Hypoxia-asphyxia group(n=6) breathed in 8% oxygen for 3 hours and then were suffocated with nitrogen;asphyxia group(n=6) were suffocated with nitrogen;but controls(n=6) were killed by decapitation.All rats′brains were isolated for determining the PDE activities by HPLC. Results PDE activities in hypoxia-asphyxia group〔(230±28) nmol·g-1·min-1〕 were lower than those in controls 〔(365±67) nmol·g-1·min-1〕(P<0.01); but PDE activities in asphyxia group〔(439±138) nmol·g-1·min-1〕 were higher than controls (P>0.05). Conclusion There exists a phase-change for the PDE activities in rats′ brain during hypoxia,PDE may participate in the pathological course of brain injury during hypoxia.
