补气中药对正常及辐射小鼠脾脏巨噬细胞的影响

目的:通过观察补气中药人参、黄芪、白术、西洋参对巨噬细胞的影响,探讨其影响机体免疫应答过程的可能作用机制。方法:运用流式细胞仪观察分析正常小鼠及辐射损伤小鼠经口服给予补气中药后,脾脏巨噬细胞的比例变化和巨噬细胞表面主要组织相容性抗原分子MHC-I、MHC-II的表达强度;采用酶标仪检测、观察分析补气中药对辐射损伤小鼠脾脏巨噬细胞色氨酸含量的影响,以了解对IDO酶活性的影响。结果:补气中药均可提高正常小鼠脾脏巨噬细胞的比例,尤以西洋参显著;并可增加辐射损伤小鼠脾脏巨噬细胞的比例。人参、黄芪、白术可下调巨噬细胞表面的MHC-I类分子,但可上调MHC-II类分子的表达,西洋参则对MHC-I、MHC-II类分子表达呈下调。经抗原刺激后,补气中药都提高巨噬细胞色氨酸含量,西洋参作用较显著。结论:人参、黄芪、白术、西洋参虽然同为补气中药,但其对巨噬细胞的影响显示出一定差异。

Observation on Effect of Qi-invigorating Herbs on the Function of Macrophage in Spleen in the Normal and Radiated Mice

Objective: To approach the possible mechanism of action on immune response through observation on the effects of radix ginseng,radix astragali,rhizoma atractylodis macrocephalae,and radix quinquefolii on macrophage function. Methods:The ratio variation of splenic macrophage and the expression intensity of the major histocompatibility antigens MHC- I and MHC-II on the macrophage surface were determined by flow cytometer,and the enzyme activity of IDO were expressed through typtophan content determined by enzyme analysis meter. Results:Qi-invigorating herbs increased the ratio of splenic macrophage in normal and radiated mice,especially radix quinquefolii. Radix ginseng,radix astragali,rhizoma atractylodis down-regulated the expression of MHC- I,and up-regulate the expression of MHC-II. Radix quinquefolii down-regulated the expression of MHC-I and MHC-II. Qi-invigorating herbs increased the typtophan content of macrophage after antigenic stimulation,especially radix quinquefolii. Conclusion:These qi-invigorating herbs have different effects on macrophage.
