流感病毒A单抗杂交瘤细胞的制备及鉴定

目的制备抗A型流感病毒的单克隆抗体，用以建立特异、灵敏、简便的A型流感金标试纸检测方法。方法将A型流感病毒为完全抗原免疫BALB／c小鼠，得到的脾细胞与SP2／0细胞融合，获得稳定分泌抗流感A单抗的杂交瘤细胞株，对抗体进行鉴定。结果用杂交瘤技术，得到了3株可稳定分泌抗流感A单抗的杂交瘤细胞株，能分泌高滴度的针对A型流感病毒的特异单抗。结论本研究建立了抗A型流感病毒杂交瘤细胞株单克隆抗体，为金标法快速诊断A型流感感染打下了试验基础。

Preparation and identification of monoclonal antibody for Influenza A virus

Objective To prepare the McAbs for Influenza A virus and set up a specific, sensitive and simple method marked by gold to detect Influenza A virus. Methods Influenza A virus was used for the immunization of BALB/c mice. The hybridoma cell strains stably secreting McAbs against Influenza A virus was obtained by fusion of murine splenocytes with SP2/0 plasmacytoma cells, and the secreted McAb was characterized. Results Three hybridoma cell strains which can steadily secrete specific McAbs was obtained. Conclusion The McAbs established by our method could be highly specific for Influenza A virus, which provides a basic condition for development of rapid diagnosis marked by gold to detect Influenza A virus.
