摘    要:
目的建立测定犬血清中盐酸伪麻黄碱的方法.方法采用高效液相色谱法,血清样品用乙醚提取,以ODS C18为固定相,甲醇:乙腈:磷酸盐缓冲液(pH 3.5)=30:10:60为流动相,UV 210 nm紫外检测.结果在0.031252.0 mg·L-1浓度范围线性良好,最小检测浓度为0.03125 mg·L-1,最低检测限为0.625 ng.其日内差(n=5)及日间差(n=5)RSD分别小于7.5%和7.6%,相对回收率(n=5)大于91.0%.结论方法简便灵敏,为盐酸伪麻黄碱的药代动力学研究提供了检测方法.

Abstract:
AimTo establish a method for determination of pseudoephedrine hydrochloride in dog serum.MethodsPseudoephedrine was extracted with ethyl ether,and then measured by RP-HPLC using a ODS C_(18)coWlumn asstationary phase and methanol-acetonitrile-phosphate buffer solution(pH 3.5)(301060)as mobile phase with ultraviolet Absorption at 210nm.ResultsThe method proved to be linear in the range of 0.03125~2.0 mgL~(-1)and the lower limit of detection of pseudoephedrine hydrochloride in serum was 0.03125 mgL~(-1).The RSD was under 7.5%(n=5)and 7.6%(n=5)for inter-assay and intra-assay respectively,and the relative recovery rate was superior to 91.0%.ConclusionThis method is sensitive,specific,simple and validated for use in pharmacokinetic study.
