留兰香油对脂多糖引起的大鼠肺气肿样改变及对MMP-9表达的影响

摘    要:目的:观察留兰香油对脂多糖(LPS)所致大鼠肺气肿样改变及其对肿瘤坏死因子-α(TNF-α)、白细胞介素一1β(Ⅱ,1β)和组织抑制因子-1(TIMP-1)含量及基质金属蛋白酶-9(MMP-9)表达的影响.方法:反复气道内滴入脂多糖8周建立大鼠肺气肿样改变模型,设立正常对照组、模型组、地塞米松组及留兰香油10,30,100 mg·kg-1组,各给药组灌胃治疗4周.计数大鼠支气管肺泡灌洗液(BALF)中自细胞总数及其分类;肺组织病理切片行HE,AB-PAS染色,观察肺泡结构、气道炎症及杯状细胞化生,并对病理图像进行形态学测量分析;测定肺组织TNF-α,IL-1β和TIMP-1的含量及MMP-9的表达水平.结果:留兰香油10,30,100 mg·kg-1组及地塞米松组能明显减少LPS引起的肺泡破坏;留兰香油100 mg·kg-1组及地塞米松组能明显降低BALF中自细胞总数及分类中的中性粒细胞、淋巴细胞数量并抑制杯状细胞化生;地塞米松对TNF-α,IL-1β,TIMP-1及MMP-9的表达均有抑制作用;留兰香油30,100 mg·kg-1能分别降低肺组织匀浆中的TNF-α,IL-1β含量;留兰香油10,30,100 mg·kg-1对TIMP-1的表达无影响,却能显著降低肺组织中MMP-9的表达水平.结论:留兰香油对脂多糖引起的大鼠肺气肿样改变有一定的保护作用,其机制可能是留兰香油降低了肺组织TNF-α及IL-1β的含量,抑制了MMP-9的过表达.
