摘 要: | 目的:探讨康复期强制戒毒人员淋巴细胞基因组DNA甲基化水平和T细胞亚群间的相关性。方法:抽提康复期强制戒毒人员和正常人淋巴细胞基因组DNA,采用整体甲基化定量试剂盒检测基因组DNA甲基化水平;T细胞亚群检测采用流式细胞术。结果:分别对80例康复期强制戒毒人员和正常健康人群进行淋巴细胞基因组DNA甲基化水平检测发现,康复期强制戒毒人员淋巴细胞基因组DNA甲基化百分率为(15.37±3.28)%,显著低于正常健康人群的(23.18±7.98)%,差异具有统计学意义(P〈0.05)。康复期强制戒毒人员CD3+、CD3++CD4+和CD3++CD4+/CD3++CD8+均明显低于正常健康人群(P〈0.05)。且CD3+、CD3++CD4+与淋巴细胞基因组DNA甲基化水平呈正相关(r=0.21,0.36,P〈0.05)。结论:康复期强制戒毒人员淋巴细胞基因组DNA甲基化水平显著低于正常人水平,其淋巴细胞基因组DNA甲基化水平和CD3+、CD3++CD4+明显相关。

Abstract: | Objective: To detect the correlation between the genomic DNA methylation of lymphocyte and the change of T lymphocyte subset among drug users. Method: Genomic DNA was extracted from lymphocytes of drug users and healthy people ( control ) . And DNA methylation levels was detected by Global DNA Methylation Quantification Ultra Kit. T-lymphocyte subsets of peripheral blood were measured by flow cytometer ( FCM ) . Result: 80 drug users and 80 healthy people were involved in our study. DNA methylation levels in lymphocytes of d~g users was 15.37%, which was significantly lower than that of control ( 23.18%, P〈0.05 ) . The levels of CD3+, CD3++CD4+ and CD3++CD4+/CD3++CD8+ reduced significantly in the drug users compared with the healthy control group ( P〈0.05 ) . The DNA methylation levels showed the positive correlation with the levels of CD3+, CD3++CD4+ ( r=-0.21, 0.36, P〈0.05 ) . Conclusion: The methylation level of genomic DNA in lymphocyte of drug users is significantly lower than that of healthy people, and The DNA methylation levels shows the positive correlation with the levels ofCD3+, CD3++CD4+. |
