化浊解毒活血通络法对脑缺血再灌注损伤小鼠行为学及海马神经细胞形态学的影响

摘    要:目的探讨化浊解毒活血通络法对脑缺血再灌注损伤的作用机制。方法将50只昆明小鼠随机分为假手术组、模型组、中药高剂量组、中药低剂量组、尼莫地平组,每组10只。采用线栓法制造小鼠右侧大脑中动脉脑缺血再灌注模型。手术造模成功后假手术组、模型组灌服生理盐水20ml/(kg.d),尼莫地平组灌服尼莫地平液19.6g/(kg.d),中药高剂量组、中药低剂量组分别给予化浊解毒活血通络中药19.6g/(kg.d)及9.8g/(kg.d)灌服。均每日1次,14天后进行行为学测试,并通过光镜观察小鼠海马CA1区的病理形态学改变。结果与模型组相比,中药高剂量组能提高脑缺血再灌注损伤小鼠的学习和记忆成绩,改善小鼠海马CA1区的神经细胞形态学表现,并且优于尼莫地平组和中药低剂量组(P<0.05)。结论化浊解毒活血通络法对脑缺血再灌注损伤小鼠的行为学能力及海马CA1区的神经细胞具有保护作用,这可能是该法对抗脑缺血再灌注损伤的机制之一。
