多孔双相陶瓷复合重组人骨形态发生蛋白-2和碱性成纤维细胞生长因子缓释微球的生物相容性

摘    要:目的 观察复合重组人骨形态发生蛋白-2(rhBMP-2)和碱性成纤维细胞生长因子(bFGF)缓释微球的多孔双相陶瓷(BCP)组织相容性.方法 A组(rhBMP-2+bFGF缓释微球/BCP)、B组(单纯BCP)、C组(bFGF缓释微球/BCP)、D组(rhBMP-2缓释微球/BCP)和E组(不含BCP).其中bFGF、dlBMP的浓度各为5、15μg,材料均为5 mm×5 mm×1mm.采用体外细胞培养的方法测定细胞黏附能力、浸提液对于兔骨髓基质细胞(BMSCs)增殖和成骨分化的影响及体外诱导BMSCs成骨分化的能力.结果 细胞在材料上黏附、生长良好.各组噻唑蓝(MTT)吸光度值、碱性磷酸酶(ALP)值均随培养时间的延长而增大.A组与B、D和E组比较对MSC有显著的促增殖作用,但低于C组(P<0.05).A组ALP活性显著高于B、C和E组,但低于D组(P<0.05).结论 复合rhBMP-2和bFGF缓释微球的BCP具有良好的生物相容性.

Abstract:Objective To investigate the biocompatibility of biphasic ceramics phosphate (BCP) combined with recombinant human bone morphogenetic protein-2 ( rhBMP-2 ) and basic fibroblast growth factor (bFGF) microspheres. Methods The rabbits were divided into group A (rhBMP-2 +bFGF micro-sphere/BCP) , group B ( BCP), group C ( bFGF microsphere/BCP), group D ( rhBMP-2 microsphere/ BCP) and group C (The cells were seeded into culture plates). The densities of bFGF and rhBMP were 5μg and 15 (xg, respectively. All the samples were 5 mm× 5 mm ×1 mm in size. The biocompatibility of the composite samples was evaluated by the assessment on the cell adhesion, proliferation, and differentiation, using the co-cultured rabbit bone marrow mesenchymal stem cells (BMSCs) in vitro. Results The cells could adhere to the novel composite and grow well. The absorbance (A) values measured by methyl thiazolyl tetrazolium (MTT) and alkaline phosphatase (ALP) in each group were increased gradually with the culture time. The proliferation rate of the rabbit MSC was significantly higher in group A than in groups B, D, and E, but significantly lowere than in group C (P<0.05). The ALP value was significantly higher in group A than in groups B, C and E group, but significantly lower than in group D. Conclusion BCP combined with rhBMP-2 and bFGF microspheres has a good compatibility with the rabbit BMSCs.
