WWOX 及P53基因在大肠癌中的表达及与肿瘤浸润的关系

摘    要:目的探讨WWOX(WW domain-containing oxidoreductase)基因及P53在大肠癌组织中的表达情况、相关性及与肿瘤浸润转移的关系。方法用免疫组化染色的方法分别检测WWOX及P53基因在35例大肠癌组织及14例正常大肠粘膜组织中的表达情况。结果 WWOX基因在大肠癌组织中的表达明显降低,与正常组织比较差异有统计学意义(P〈0.05)。P53(突变型)基因在大肠癌组织中的表达明显高于正常组织,二者比较差异有统计学意义(P〈0.05)。WWOX与肿瘤浸润、淋巴结转移情况密切相关(P〈0.05)。P53表达与癌组织的浸润密切相关(P〈0.05),与淋巴结转移情况无关(P〉0.05)。WWOX与P53基因的表达情况均与患者的年龄、性别无关(P〉0.05)。WWOX与P53基因的表达呈负相关(r=-0.442,P=0.008)。结论 WWOX、P53(野生型)基因同为抑癌基因,且两者之间在大肠癌的发生、浸润及转移过程中有着较为明显的协同作用,联合评价其功能具有重要的临床意义。

Abstract:Objective To investigate the relationship between the expression of tumor-associated gene WWOX (WW domain-containing oxidoreductase) and P53 in large intestine carcinoma tissue and their relationship with tumor infiltration.Methods Immunohistochemical staining method was to detect the expression of WWOX and P53 gene in 35 cases of large intestine carcinoma tissues and 14 cases of normal colonic tissues respectively.Results There was a significant difference in the expression of WWOX between large intestine carcinoma tissues and normal tissue (P〈0.05).The expression of WWOX was closely related with tumor invasion depth and lymph node metastases(P〈0.05).The expression of P53 (mutant) in large intestine carcinoma tissues was significantly higher than that in normal tissues,there was a significant difference between the two kinds of tissues(P〈0.05).The expression of P53 was closely related with tumor invasion depth(P〈0.05).The expression of P53 (mutant) in large intestine carcinoma tissues was not obviously related with lymph node metastasis (P〈0.05).The expression of the two kinds of genes had no obvious correlation with the patient's age and gender(P〈0.05).The expression of WWOX was negatively related to that of P53(mutant) in large intestine carcinoma tissues (P〈0.01).Conclusion The WWOX and P53 (wild type) genes are all tumor suppressor genes,and the two genes have obvious synergistic action in the pathogenesis,invasion and transfer process of large intestine carcinoma.Therefore,the combined evaluation for the two genes' function has important clinical significance.
