冰茶栓对骨癌痛大鼠骨溶解的影响

摘    要:目的观察冰茶栓对W256细胞诱导的骨癌痛大鼠骨溶解的影响。方法将40只雌性SD大鼠分为假手术组、模型组、邦罗力组及冰茶栓组,采用骨髓腔注射W256癌细胞复制骨癌痛模型;模型复制第13天开始,假手术组和模型组给予空白栓,邦罗力组给予邦罗力20μg/kg,冰茶栓组给予冰茶栓101mg/kg。末次给药后,对各组大鼠患肢进行X线摄片并评分以观察骨溶解情况,采用苏木精-伊红染色观察骨组织病理学变化,采用抗酒石酸酸性磷酸酶染色观察破骨细胞数量。结果冰茶栓可明显降低骨癌痛大鼠患肢X线评分(P0.01),改善肿瘤引起的骨损伤,降低骨组织破骨细胞数量(P0.01)。结论冰茶栓对W256细胞诱导的骨癌痛大鼠骨溶解具有较好的抑制作用。

Effects of Bingcha Embolus against Osteolysis in Rats with Bone Cancer Pain

Abstract:Objective To observe effects of Bingcha Embolus,a traditional Chinese medicine mainly composed of Agkistrodon halys venom,borneol,and tea leaves for cancer pain,against osteolysis in rats with bone cancer pain induced by W256 tumor cells.Methods Forty female Sprague-Dawley rats were divided into four groups: sham-operation group,model group,Bondronat treated group and Bingcha Embolus treated group.The models of bone cancer pain were induced by injecting W256 tumor cells into the rat marrow cavity.From the 13th day after operation,rats in sham-operation group and the model group were treated with blank embolus,rats in Bondronat treated group were treated with Bondronat Injection(20 μg/kg),and rats in Bingcha Embolus group were treated with Bingcha Embolus(101 mg/kg).At the end of last treatment,the affected legs of all the rats were observed with X-ray,and the X-ray signs were scored for evaluating the osteolysis.The pathological changes of bone tissues were observed by using hematoxylin-eosin staining.Osteoclasts in the affected leg bone tissues were counted by using tartrate-resistant acid phosphatase(TRAP) staining.Results Bingcha Embolus significantly reduced the X-ray scores of affected legs of rats with bone cancer pain(P0.01),alleviated the destruction of bone tissues caused by W256 tumor cells,and notably reduced the number of osteoclasts in the bone tissues(P 0.01).Conclusion Bingcha Embolus has a good effect against osteolysis in rats with bone cancer pain induced by W256 tumor cells.
