人雪旺氏细胞分泌神经营养因子浓度的测定

摘    要:
目的测定体外培养的人雪旺氏细胞分泌神经营养因子在各时间点的浓度并观察其时间-浓度关系。方法取人雪旺氏细胞进行体外培养,在培养后的第1天、第3天、第7天、第14天、第21天及第28天六个时间点用间接酶联免疫吸咐法测定培养液中碱性成纤维细胞生长因子(bFGF)、睫状神经营养因子(CNTF)和脑源性神经营养因子(BDNF)的浓度。结果在人雪旺氏细胞培养后的第1天、第3天、第7天、第14天、第21天及第28天时,在培养液中均可检测到bFGF、CNTF和BDNF的存在。在培养后第21天及第28天时,培养液中三种神经营养因子的浓度都显著增加。bFGF的浓度显示出高于CNTF和BDNF浓度的趋势(P<0.01)。结论培养的雪旺氏细胞能分泌神经营养因子,其浓度随培养时间的推移而升高。

Abstract:
Objective To assay the concentration of neurotrophic factors secreted by human schwann cells that were cultured in vitro and to observe the time-concentration relationship.Methods Human schwann cells were cultured in vitro and the concentrations of basic fibroblast growth factor (bFGF), ciliary neurotrophic factor (CNTF) and brain-derived neurotrophic factor (BDNF) in the medium were assayed using an indirect enzyme-linked immunosorbent assay at 1, 3, 7, 14, 21 and 28 days after the cells were cultured.Results bFGF, CNTF and BDNF were found in the culture medium of human schwann cells at 1, 3, 7, 14, 21 and 28 days after the cells were cultured. Concentrations of the three neurotrophic factors in the culture medium significantly increased 21 and 28 days after the cells were cultured. The concentration of bFGF tended to be higher than concentrations of CNTF and BDNF (P<0.01).Conclusion Cultured schwann cells can secrete neurotrophic factors and the level of these concentrations increased in the culture medium as time passed.
