TRAIL基因对人白血病Jurkat细胞生物力学特性的影响

摘    要:
本实验研究将人类肿瘤坏死因子相关凋亡配体 (TNF relatedapoptosisinducingligand)的cDNA序列作为目的基因导入RevTet On后 ,利用在Jurkat中所建立起的四环素调控TRAIL基因表达系统 ,作为研究肿瘤细胞凋亡前后生物流变学特性变化的细胞模型。我们通过测量用强力霉素诱导TRAIL基因表达前后细胞生物力学特性的变化 ,研究TRAIL基因对Jurkat细胞生物力学特性的影响。结果显示TRAIL基因的表达对Jurkat细胞有明显的促凋亡作用。TRAIL基因表达后 ,细胞表面电荷密度减少 ,流动性下降。而且 ,本实验表明 ,TRAIL基因诱导的细胞凋亡引起了Jurkat细胞膜的二级结构改变

Abstract:
Object: to study the influence of TRAIL gene on biomechanical properties of human leukemic cell line Jurkat. Methods: We cloned the cDNA fragment of human TRAIL (TNF related apoptosis inducing ligand) into RevTet On. Making use of the TRAIL gene expression system in Jurkat as a cell model, we studied the influence of TRAIL gene on Jurkat through measuring changes of cellular biomechanical properties before and after the TRAIL gene expression, which was induced by adding teracyeline derivative doxycycline (Dox). Results: the results indicated that the TRAIL gene led to significant apoptosis. The surface charge intensively decreased, and the cell stiffness increased after the expression of TRAIL gene. Moreover, it is suggested that the apoptosis, which is induced by TRAIL gene. changes the second structure of Jurkat cell membrane. Conclusion: TRAIL gene can change the microstructure of cell membrane thus induce the impairment of cellular rheological properties.
