转铁蛋白与Bel-7402肝癌细胞耐药性关系的研究

摘    要:
目的 研究转铁蛋白与Bel-7402肝癌细胞耐药性的关系.方法 根据已知转铁蛋白序列设计引物扩增基因片段,荧光定量PCR检测其在敏感Bel-7402细胞株、阿霉素抗性Bel-7402细胞株之间的表达水平差异.结果 成功扩增Bel-7402肝癌细胞转铁蛋白基因片段,荧光定量PCR表明转铁蛋白基因在阿霉素抗性Bel-7402细胞株的表达量是敏感Bel-7402细胞株4.83倍.结论 转铁蛋白基因对肝癌细胞的耐药发生发展存在一定的作用,可为研究肝癌耐药性的备选基因,以及为肝癌耐药性检测提供新的靶标.

The relationship between transferrin and drug resistance of BEL-7402 hepatocellular carcinoma cell line

Abstract:
Objective To study the relationship between transferrin and drug resistance of BEL-7402 hepatocellular carcinoma cell line.Methods The primers for amplification of transferrin gene fragment on the basis of known gene sequence were designed,fluorescence quantitative PCR was used for detection of expression level difference between sensitive BEL-7402 hepatocellular carcinoma cell line and adriamycin resistant BEL-7402 hepatocellular carcinoma cell line.Results The transferrin gene fragment was cloned successfully and FQ-PCR proved the transferrin gene expression level of adriamycin resistant BEL-7402 hepatocellular carcinoma cell line was 4.83 times higher than that of sensitive BEL-7402 hepatocellular carcinoma cell line.Conclusion The transferrin gene presents a certain role in drug resistance development mechanism of hepatocellular carcinoma cell line,and it can be used as a new resistance gene and drug resistance detection target.
