川芎HPD-100大孔树脂和醇沉纯化物指纹图谱研究

摘    要:
目的:建立川芎HPD-100大孔树脂纯化物及醇沉纯化物的指纹图谱,筛选较优纯化方法并为有效控制其质量提供可靠方法.方法:高效液相色谱法,Phenomenex Hyperclone BDS C18(4.00 mm×250 mm,5μm),甲醇-1%冰乙酸为流动相,梯度洗脱(0~35 min,20∶80~45∶55,35~55 min,45∶55~70∶30,55~70 min,70∶30~ 80∶20),322 nm检测.结果:川芎HPD-100大孔树脂纯化物共有22个共有峰,川芎醇沉纯化物共有20个共有峰,分离好,相似度均在0.98以上,重复性好.结论:川芎采用HPD-100大孔树脂纯化较好.本方法科学、准确、专属性好,可为川芎药材及含川芎的中成药质量控制提供可靠的科学依据.

Abstract:
Objective:To establish the HPLC fingerprint of Chuanxiong Intermediate and provide a method to control the quality of Chuanxiong.Method:HPLC method was applied for quality assessment of Chuanxiong Intermediate.HPLC analysis was performed on a Phenomenex Hyperclone BDS C18(4.00 mm×250 mm,5 μm) by gradient elution using methanol-1% acetic acid solution as mobile phase.Flow-rate was 1 mL·min-1.The detection wavelength was at 322 nm.Result: The intermediate of Rhizoma Chuanxiong were closely related to each other.Conclusion: The method is rapid and accurate.It was used for determination of Chuanxiong Intermediate and Chinese formulated medicine with it.
