溶血卵磷脂在胰性脑病发病机制中的作用

摘    要:目的 探讨溶血卵磷脂(LPC)在胰性脑病(PE)发病机制中的作用环节和效应机制。方法 将SD大鼠随机分组,用AhoHJ法将实验组、对照组1制成急性水肿型胰腺炎模型,随后对实验组大鼠尾静脉应用LPC ;对照组1从尾静脉注入生理盐水;对照组2是假手术组,经尾静脉注入LPC。连用LPC 5d后的7~10d ,应用辣根过氧化物酶 二氨基联苯氨(HRP DAB)显色法检测其血脑屏障(BBB)是否开放,同时行脑组织切片丽春红、固蓝染色观察有无脱髓鞘改变。结果 实验组大鼠的BBB开放较明显,对照组的BBB几乎无开放。实验组脱髓鞘较明显,对照组脱髓鞘不明显,差异均有统计学意义(P <0 .0 5 )。结论 LPC对胰腺炎大鼠的BBB具有开放作用,对胰腺炎大鼠脑部有脱髓鞘作用;LPC是PE的致病因素之一,在PE发病机制中起一定作用

Abstract:Objective To explore some certain processes and effects of lysolecithin choline (LPC) in the pathogenesis of pancreatic encephalopathy (PE).Methods SD rats were divided into test group,control group 1 and control group 2 at random.Acute edematous pancreatitis models were induced according to the Aho HJ method for test group and control group I.The rate in test group were subjected to the intravenous injection of LPC,while those in the control group I to saline via tail vein.The rats in control group 2 were subjected to the intravenous infection of LPC with pseudo-operation.Horseradish peroxidase (HRP) was used as a tracer to determine if the blood-brain barrier (BBB) was open 7-10 days after successive application of LPC for 5 days.The extravasated tracer was showedby diaminobenzidine (DAB).Rats' brain tissue sections were examined by ponceau stain and Luxol Fast Blue stain to determine whether the brains were demyelinated at the same time.Results In test group,the BBB permeability of the rats was increased greatly and obvious demyelination was observed in brains,while in both control groups the rats had basically intact BBB and scarce demyelination.There was statistical difference between the test group and control groups.Conclusion LPC increase the BBB permeability and demyelinates the brain on pancreatitis models,suggesting that LPC might play an important role in the pathogenesis of PE.
