三氧化二砷对肝门部胆管癌细胞增殖及凋亡的影响

摘    要:目的利用人肝门部胆管癌细胞系(FRH-0201)接种裸鼠,建立肝门部胆管癌原代移植瘤模型;研究三氧化二砷(As2O3)对肝门部胆管癌细胞增殖及凋亡的影响。方法将FRH-0201细胞系(120代)接种于50只Balb/c裸小鼠脾脏,建立肝门部胆管癌原代移植瘤模型。荷瘤裸鼠分为未用药组和三氧化二砷组。用药后观察各组裸鼠移植瘤体积变化;采用原位末端标记法检测肿瘤组织细胞凋亡;利用免疫组化观察移植瘤中核增殖抗原(PCNA)的表达。结果建立了肝门部胆管癌原代移植瘤模型;三氧化二砷组对原代移植瘤瘤体体积具有抑制作用;三氧化二砷组能诱导细胞凋亡,抑制癌细胞增值,降低PCNA的表达,与未用药对照组相比差异显著(P〈0.05)。结论在原代人肝门部胆管癌动物模型中.三氧化二砷能缩小肿瘤体积,诱导肿瘤细胞凋亡,并能降低肿瘤细胞的PCNA表达。

Abstract:OBJECTIVE To establish a tumor model in nude mice by splenic inoculation of hilar cholangiocarcinoma cell line. To study the effects of As2O3 on tumor model of hilar cholangiocarcinoma. METHODS Human hilar cholangiocarcinoma cell line(FPH-0201 cells)was established and cultured with a fresh human hilar cholangiocarcinoma specimen, thus establishing tumor model of hilar cholangiocarcinoma. The cancer bearing nudc mice of hilar cholangiocarcinoma were divided into 2 groups:control group (no treatment) and As2O3 group. After treated, the volume change of tumor were observed. TUNEL were performed to examine apoptosis. The expression of PCNA were detected by immunohistochemical. RESULTS We established a tumor model in nude mice of hilar cholangiocarcinoma cell line. The volume of tumor were significantly decreased in As2O3 group compared with control group(P 〈 0.05). The As2O3 could produce a stronger apoptotis effect on tumor cells than control group(P 〈 0.05), as well as depress expression of PCNA. CONCLUSION As2O3 can inhibit the growth of tumor, induce the apoptosis of tumor, inhibit the expression of PCNA.
