大鼠烫伤创面愈合过程中皮肤基底膜层黏连蛋白的变化

摘    要:目的:研究烫伤愈合过程中层黏连蛋白(LN)的表达。方法:采用免疫组化方法在大鼠浅Ⅱ度烫伤(伤后1,3,5,7,10,14d组)、Ⅲ度烫伤切痂混合移植模型(大张异体皮移植3,5,7,14,21,28d组)上研究了愈合过程中LN的变化。结果:正常皮肤基底膜中含有LN,而创伤后表皮细胞迁移缘LN消失,愈合后表皮下LN又出现。混合移植后7d出现异体皮排异反应,表现为异体表皮细胞铲除,而异体表皮下仍可见LN阳性表达。异体皮移植后10d,从自体皮迁移来的表皮细胞下有零星LN,而异体表皮细胞被完全铲除,异体皮基底膜处LN仍基本保持完整。异体皮移植后21d,自体表皮已基本覆盖异体皮区,此时LN表达已基本完整。结论:烫伤愈合过程中LN的变化有助于创面愈合。

Abstract:Objective To study the expression changes of laminin in the process of the wound healing. Methods The expression changes of laminin were observed by the immunohistochemical method in rats with experimental paritial thickness scald (post injury 1, 3, 5, 7, 10, 14 day groups) and extensive full thickness burn following intermingled transplantation ( a large sheet of allograft inlaid with small stamps of autoskin 3, 5, 7, 14, 21, 28 day groups). Results Laminin existed in the normal basement membrane of skin and disappeared in the epidermic cell transplantation layers posterior to injury. But it emerged again after wound healing. The rejection of alloepidermis occurred at the 7th day after intermingled transplantation, which characterized by evacuation of the heterogeneous epidermic cells and a positive expression of laminin in the heterogeneous subepidermis. Ten days after transplantation, episodic laminin could be seen in the autogeneous subepidermis, but the heterogeneous subepidermic cells were absolutely evacuated. In the meantime, the basement membrane of skin grafting was completely formed. On the 21st day after transplantion, the autogeneous subepidermis almost covered all the heterogeneous subepidermic region combining with a nearly complete expression. Conclusions The changes of the expression of laminin are beneficial to the wound healing. The allografts in intermingled grafting may provide the component of basement membrane of skin for the healing of intermingled transplantation.
