宫清方诱导T细胞亚群偏移减轻药物流产小鼠子宫出血的实验研究

摘    要:目的:探讨宫清方诱导T细胞亚群偏移减轻药物流产后子宫出血的作用与机制。方法:孕鼠40只,随机分为米非司酮(RU486)组1.5 mg/(kg.d)-1]及宫清方小77.04 mg/(kg.d)-1]、中154.08 mg/(kg.d)-1]、大308.16 mg/(kg.d)-1]剂量组,各10只。改良碱性正铁血红素法检测子宫出血量,流式细胞术检测母胎界面Th1(CD3+CD4+IFNγ+)、Th2(CD3+CD4+IL-4+)、Th17(CD3+CD4+IL-17A+)、Treg(CD4+CD25+Foxp3+)亚群比例,RT-PCR法检测小鼠子宫组织干扰素γ(IFNγ)、白细胞介素-4(IL-4)、白细胞介素-17A(IL-17A)、转化生长因子-β(TGF-β)mRNA转录水平。结果:宫清方作用后,药物流产小鼠子宫出血量明显减少(P<0.05),Th1、Th17亚群比例(P<0.05)及Th1/Th2、Th17/Treg比值升高(P<0.05),Th2、Treg亚群比例降低(P<0.05),IFNγ、IL-17A mRNA表达升高,IL-4、TGF-βmRNA表达降低。结论:宫清方能优势诱导药物流产小鼠T细胞向Th1、Th17亚群偏移,进而有效减少药物流产后子宫出血。

Abstract:Objectives:To explore the mechanism underlying the bleeding reducing effect of Gongqing decoction via regulating T-cell subsets.Methods:Total 40 pregnant mice were assigned into mifepristone(RU486) group(1.5 mg/(kg·d)-1),Gongqing decoction low-dose77.04 mg/(kg·d)-1],middle-dose154.08 mg/(kg·d)-1] and high-dose308.16 mg/(kg·d)-1] groups randomly(n=10 for each).The volume of uterine hemorrhage was measured by the method of alkaline hematin photometric.The percentages of Th1 subset(CD3+CD4+IFNγ+),Th2 subset(CD3+CD4+IL-4+),Th17 subset(CD3+CD4+IL-17A+) and Treg subset(CD4+CD25+Foxp3+)in mice uterine tissue were detected by flow cytometry.The mRNA expressions of IFNγ、IL-4、IL-17A、TGF-β were analyzed using RT-PCR.Results:Compared to RU486 group,the uterine bleeding volume reduced significantly in Gongqing decoction groups.The percentages of Th1 and Th17 cells increased while the percentages of Th2 and Treg cells decreased after Gongqing decoction administration(P<0.05).Additionally,Gongqing decoction up-regulated IFNγ and IL-17A mRNA expression and down-regulated IL-4 and TGF-β mRNA expression in a dose-depended manner(P<0.05).Conclusion:Gongqing decoction reduced the uterine bleeding volume of RU486-induced abortion mice efficiently via promoting Th1 and Th17 skews.
