摘 要: | 目的 探讨盐酸多奈哌齐(Done)对戊四氮(PTZ)点燃慢性癫痫大鼠的认知功能损害的干预作用.方法 60只雄性SD大鼠随机分为盐水对照组(n=18)和癫痫模型组(n=42),模型组以PTZ腹腔注射(35mg·kg-1·d-1)21 d造成慢性癫痫大鼠模型后,再随机分为PTZ组和Done组,2组继续给予PTZ注射的同时,Done组给予Done(1 mg/kg)灌胃21 d,停药后采用Morris水迷宫、跳台和事件相关电位P300的方法观察PTZ点燃癫痫模型学习记忆功能的改变、脑电生理的变化以及Done的影响作用.结果 PTZ组水迷宫中逃避潜伏期延长(F=29.8,P=0.005),平台象限游泳时间减少(t=5.32,P=0.008),穿越平台次数减少(t=17.5,P=0.03);跳台试验中反应时间延长(t=2.49,P=0.04),步下潜伏期缩短(t=3.86,P=0.02),错误次数增加(t=24.1,P=0.04);P300峰潜伏期延长(t=3.11,P=0.03).在水迷宫和跳台试验的测试初期,Done组与对照组相比有明显差异(P<0.05),但在测试后期无明显区别,P300峰潜伏期无差异.结论 PTZ点燃的慢性癫痫大鼠有空间学习记忆能力等认知功能障碍,Done可改善慢性癫痫大鼠空间学习能力和提高认知功能.

Effects of donepezil on the cognitive function of chronic epileptics rats kindled by pentylenetetrazole |

Abstract: | Objective To explore the effect of donepezil(Done)on the spatial learning and memory capacity of chronic epileptics model rats kindled by pontylentetrazole(PZT).Methods Sixty male Sprague-Dawley rats were divided randomly into control group(n=18),in which rats received intraperitoneal injections of the 0.9%0.9% saline) for 21 days.After completely kindled.model group rats divided randomly into PTZ group and Done group.Donepozil was orally administrated for 21 days in the Done group,meanwhile,PTZ was applied sequentially in PTZ group and Done group.After the last application of donepezil.cognitive function was tested by Morris water maze,step-down test,and event-related potential P300.Results The escape latency,total swimming time in platform quadrant and crossing times in the Morris water maze test were different significantly when compared PTZ group with control group (F=29.8,P=0.005;t=5.32,P=0.008;t=17.5,P=0.03,respectively),similarly results in the step-down test (t=3.86,P=0.02) and P300 peak latency were obtained(t=3.11,P=0.03).There were significantly differences in the early term of water maze and step-down test when Done group compared with control group (P<0.05),however,no differences were found in the late term of water maze and step-down and P300 test.Conclusion PTZ kindled rats are cognition impairment.and donepezile can improve the spatial learning capacity partially,elevate the spatial memory capacity significantly and prolong the P300 latency,thus boost the cognitive function of chronic epileptics rat. |
