硫氧还蛋白对缺氧/复氧大鼠海马神经元caspase-3活性的影响

摘    要:目的:探讨硫氧还蛋白(Trx)和caspase-3在鼠海马神经元缺氧/复氧损伤中的作用。方法:原代培养10d的新生鼠海马神经元缺氧3h复氧12h,24h和48h,分为正常对照组、缺氧/复氧组和Trx组,用MTT比色法测定各时点的神经元存活率;底物裂解法检测caspase-3活性。结果:缺氧/复氧组海马神经元复氧12-48h,海马神经元caspase-3样蛋白酶活性明显增高,以复氧24h为最高,而存活率进行性下降;Trx组caspase-3样蛋白酶活性曲线与缺氧/复氧组相似,但明显下降,细胞存活率较缺氧/复氧组明显提高。结论:海马神经元缺氧/复氧后,caspase-3样蛋白酶被激活,参与神经元损伤过程;Trx对神经元有保护作用。

The Effect of Thioredoxin on Caspase-3 in Rat Hippocampal Neurons by Hypoxia/Reoxygenation

Abstract:Objective To study the roles of thiorodoxin and caspase-3 in rat hippocampus neurons following hypoxia/reoxygenation. Methods Ten-day-old neuronal cells were divided into three groups, the rat hippocampal neurons were induced hypoxia for 3 hours, followed by 12 to 48 hours of reoxygenation in two groups, and one being pretreated with thioredoxin for 1 hour; the left group was control group. The neuronal viability was estimated by MTT assay, and caspase-3 activities were measured by a colorimetric method. Results The caspase-3 was activated in rat hippocampal neurons during hypoxia/reoxygenation, and caspase-3 like activity was significantly increased after reoxygenation, with the peak at 24 hours of reoxygenation. Decreased caspase-3 like activity was detected in the neurons pretreated with thioredoxin. On the other hand, the neuronal viability was decreased in H/R group, but was improved in thioredoxin group. Conclusion caspase-3 contributes to the neuronal damage following hypoxia/reoxygenation; and thioredoxin has the neuroprotective effects.
