| 摘 要： | 本文以提纯冻存抗原和回收抗原包被40孔聚苯乙烯固相载体,采用酶联免疫吸附试验法(ELISA)检测抗可溶性抗原杂交瘤上清液。比较了不同的封闭剂及其浓度对实验结果的影响。实验结果表明,应用回收抗原和提纯的抗原检测结果一致,可代替首次抗原进行检测,并节约试剂的用量。同时证明20％小牛血清-吐温磷酸缓冲液(FCS-PBS-T)作为封闭剂检测,效果较好,明显优于用1％BSA-PBS-T作为封闭剂。

| Abstract: | The hybridoma supernatant against a Soluble antigen was added into 40-well polystyrene microtiter plate coated with purified and recovered antigen and measured by Enzyme-linked Immunosorbent Assay(ELISA).The effect of different blockings and their concentrations were compared with each other.The result indicated that the recovered antigen had the same effect as the purified antigen.Furthermore,the former could substitute for the latter in order to save purified antigen.We also observed that 20%FCS-PBS-T as a blocking had an advantage over 1%BSAPBS-T |
