雷公藤多甙及雌孕激素治疗对大鼠卵巢Bcl-2和Bax表达的实验研究

摘    要:目的探讨雷公藤多甙(TWP)致雌性大鼠生殖系统损害机制及防治方法。方法取50只SD雌性大鼠随机分为对照组,雌孕激素干预组(干预组),雌孕激素治疗组(治疗组),恢复组和雷公藤组。运用放免法测定血清性激素水平的变化,HE染色观察卵巢组织变化,免疫组化染色法检测卵巢组织颗粒细胞凋亡蛋白Bcl-2和Bax的表达水平。结果与对照组相比,雷公藤组和恢复组大鼠血清黄体生成素(LH)和卵泡刺激素(FSH)水平明显升高,雌激素(E2)的分泌明显下降,且差异有统计学意义(P值均〈0.05);卵巢组织明显受损,各级卵泡明显减少,卵泡闭锁较多,干预组各指标则无明显改变;大鼠颗粒细胞凋亡蛋白Bcb2的表达在雷公藤组明显低于对照组,而干预组及治疗组均高于雷公藤组;Bax蛋白在各组的表达则相反(P〈0.01),其中干预组与对照组无明显差异(P〉0.05)。结论TWP可致雌性大鼠生殖系统的损害,雌孕激素替代治疗对卵巢功能有较好的保护作用,其机制可能与Bcl-2蛋白表达上调以及Bax表达下凋有关。

Effects of estrogen-progestogen replacement therapy and tripterygium wilfordii polyglycosidium on expressions of Bcl-2 and Bax in ovary cells of rats

Abstract:Objective To explore the protective effects of estrogen-progestogen replacement therapy (HPT) on impaired genital system induced by tripterygium wilfordii polyglycosidium (TWP) in female rats and its mechanism. Methods 50 SD female rats were randomly divided into five groups: control group, intervention group, treatment group, recovery group and TWP group. The levels of serum gonadal hormone were detected by radioimmunoassay. The histological changes of the ovary were observed after HE staining. Using the immunohistochemical staining technique we detected the expression levels of apoptosis regulatory protein Bcl-2 and Bax in ovarian granular cell. Results The level of E2 secreted by ovary was lower and the levels of LH and FSH secreted by pituitary were higher in TWP group and recovery group than those in control group(P〈0.05) and there were no significant differences in levels of these three hormones between control group and intervention group (P 〉 0.05). In TWP and recovery group,ovarian tissues were significantly injured with obvious decreasing of ovarian follicles at different stages and more clausura of these follicles. Compared with control group, estrogen replacement therapy combined with progestogen could restrain these pathological changes induced by TWP. In TWP group, Bcl-2 expression in ovary cells of rats was lower than that in control group, but in intervention group and treatment group, it was obviously higher than that in TWP group. However,Bax expression showed the contrary changes at the same time in each group (P〈0.01) and there was no significant difference in Bax expression between control group and intervention group (P〉 0. 05). Conclusion HPT can prevent ovary injury induced by TWP. Its mechanism may be partly related to up- regulation of Bcl-2 expression and down-regulation of Bax regulation.
