异体脂肪源间充质干细胞移植后稳定嵌合体的形成及免疫耐受的诱导

摘    要:目的研究器官移植中脂肪源F1k1+CD31—CD34-细胞诱导免疫耐受的可能性。方法以CM—DiI荧光染料示踪脂肪源F1k1+CD31-CD34-细胞的体内分布情况,并以PCR方法检测Y染色体的存在以进-步鉴定;以皮肤移植实验观察脂肪源F1k1+CD31-CD34-细胞移植组小鼠对供者来源组织器官移植物的反应性。结果脂肪源F1k1+CD31-CD34-细胞可进入并较长期(30d)存在于异基因小鼠免疫器官内;在细胞移植组小鼠脾脏中可检测到供者来源T细胞,占受者脾细胞6.68%;脂肪源F1k1+CD31-CD34-细胞移植组小鼠供者来源皮肤移植物存活〉90d,未处理组平均为10d。结论异基因脂肪源F1k1+CD31-CD34-细胞移植后可形成稳定嵌和体,并诱导特异性免疫耐受的产生。
