姜黄素对高浓度七氟醚所致老年大鼠学习记忆能力改变及COX-2表达影响

摘    要:目的:通过对比分析姜黄素对高浓度七氟醚所致老年大鼠学习记忆能力改变及环氧化酶2 (Cyclooxygenase-2,COX-2)表达的影响,为预防治疗老年患者术后学习记忆能力下降提供理论依据。方法:选取无特定病原体级SD雄性大鼠48只为研究对象,随机分为三组,每组16只大鼠,分别为七氟醚组、空白对照组和姜黄素组,使用酶连免疫法检测三组大鼠海马内β-淀粉样蛋白(amyloidβ-protein,Aβ)表达水平,使用Western检测法检测三组大鼠海马内COX-2和γ-分泌酶(human beat-site app-cleaving enzy me-1,BACE-1)表达水平,记录水迷宫测试结果。结果:七氟醚组大鼠第3天、5天潜伏期明显长于对照组大鼠,5天潜伏期明显长于姜黄素组大鼠,5天总路程明显长于对照组大鼠和姜黄素大鼠,随着吸入七氟醚时间延长,七氟醚组大鼠穿越平台象限次数明显少于对照组和姜黄素大鼠,姜黄素组大鼠3天、5天潜伏期长于对照组大鼠,差异有统计学意义(P<0. 05),姜黄素组大鼠总路程及穿越平台次数与对照组大鼠相比差异无统计学意义(P>0. 05);姜黄素组与七氟醚组大鼠海马内Aβ表达水平均高于对照组大鼠,七氟醚组大鼠海马内Aβ表达水平明显高于姜黄素组大鼠,差异有统计学意义(P<0. 05);七氟醚组大鼠BACE-1表达水平高于对照组和姜黄素组大鼠,姜黄素组大鼠BACE-1表达水平高于对照组大鼠,差异有统计学意义(P<0. 05);三组大鼠COX-2表达水平无明显差异(P>0. 05)。结论:姜黄素能够降低老年大鼠海马组织内Aβ和BACE-1表达水平,减轻高浓度七氟醚对老年大鼠学习记忆能力的损害。

Abstract:Objective:By comparing and analyzing the effect of Curcumin on the learning and memory ability and the expression of cyclooxygenase-2(COX-2) for aged rats with high concentration of sevoflurane,the prevention and treatment of postoperative learning and memory ability in elderly patients provided a theoretical basis.Methods:Forty-eight SD male rats without specific pathogens were randomly divided into three groups,16 rats in each group,which were sevoflurane group,blank controlled group and Curcumin group,and were detected by enzyme-linked immunoassay.The expression levels of amyloid β-protein(Aβ) in the hippocampus of rats were detected by Western detection.The human beat-site app-cleaving enzyme-1,BACE-1) expression level was detected by Western detection.Recording water maze test results.Results:The latency of the3 rd and 5th day of the sevoflurane group was significantly longer than that of the controlled group.The 5d incubation period was significantly longer than that of the curcumin group.The 5d total distance was significantly longer than that of the controlled group and curcumin rats,with the inhalation of sevoflurane.The number of quadrants crossing the platform in the sevoflurane group was significantly lower than that in the controlled and Curcumin rats.The incubation period of the 3d and 5d rats in the Curcumin group was longer than that in the controlled group(P< 0.05).There was no significant difference in the total distance and the number of crossing platforms between the rats in the group and the control group(P>0.05).The expression of Aβ in the hippocampus of the curcumin group and the sevoflurane group was higher than that in the control group.The expression of Aβ in the hippocampus of rats in the Sevoflurane group was significantly higher than that in the curcumin group(P<0.05).The expression of BACE-1in the sevoflurane group was higher than that in the controlled group and the Curcumin group.The expression of BACE-1in the rats in the Curcumin group was higher than that in the controlled group(P<0.05).There was no significant difference in the expression of COX-2among the three groups(P > 0.05).Conclusion:Curcumin can reduce the expression of Aβand BACE-1in hippocampus of aged rats and alleviate the damage of high concentration of sevoflurane on learning and memory ability in aged rats.
