胃康舒宁对胃癌前病变大鼠胃黏膜上皮细胞凋亡及其相关调控基因表达的影响

目的:探讨胃康舒宁对大鼠胃癌前病变(PLGC)的治疗机制,为临床应用提供依据。方法:将60只Wistar大鼠随机分为4组(正常组、模型组、维酶素组和胃康舒宁组),采用饮用N-甲基-N-亚硝基-N-硝基胍(MNNG)建立大鼠胃癌前病变模型,用维酶素和胃康舒宁治疗12周后,观察各组实验大鼠胃黏膜细胞凋亡指数、Bcl-2和Bax蛋白表达情况及Bcl-2/Bax比值的变化。结果:与模型组大鼠相比,用药组大鼠胃黏膜细胞凋亡指数和Bax蛋白表达明显提高,而Bcl-2蛋白表达及Bcl-2/Bax比值显著降低,胃康舒宁组明显优于维酶素组。结论:胃康舒宁通过调节病变细胞Bcl-2和Bax蛋白的异常表达,改变Bcl-2/Bax比值,使细胞凋亡趋于正常,而对大鼠胃癌前病变具有良好的治疗和逆转作用,其疗效优于维酶素。

Effects of Weikangshu'ning on the Epithlial Cell Apoptosis and Its Regulatory Gene in Rats Gastric Mucosal with Precancer Lesions of Gastric Carcinnoma

Objective:To study the therapeutic mechanism of Weikangshu'ning on precancer lesions of gastric carcinnoma(PLGC)in rats,and offer the experimental foundation for clinical applying.Methods:60 Wistar rats were randomly divided into control group,PLGC group,vitacoenzyme group and Weikangshu'ning group.Models of PLGC were induced by N-methly-N-nitro-N-nitro-soguanidine(MNNG),gastric mucosal aoptosisindex,and the expression of Bcl-2 and Bax protein were examined in each group respectively.Results:When compared with the PLGC group,Weikangshu'ning could improve the the expression of Bax and the apoptosisindex(AI),the expression of Bcl-2 and the ratio of Bcl-2/Bax was reduced markedly.There were significant differences between the vitacoenzyme group and Weikangshu'ning group.Conclusions:Weikangshu'ning can regulate the expression of Bcl-2 and Bax protein,influence the ratio of Bcl-2/Bax,make the apoptosisindex approach control group.So it has obvious curative effects on PLGC and can take a turn for the worse.Therefore,its therapeutic effects are better than that of vitacoenzyme.
