黄体酮对大鼠脑缺血区水和钠钾钙含量的影响

摘    要:目的 证实黄体酮对缺血再灌注性脑损伤的保护作用 ,并初步探讨其作用机制。方法 制作大鼠脑缺血再灌注模型 ,将大鼠随机分为 6组 :黄体酮 (progesterone ,PROG)预防用药组、PROG预防并治疗组、PROG治疗组、单纯缺血再灌注组、阳性和阴性对照组。分别检测缺血再灌注后脑组织H2 O、Na+ 、K+ 和Ca2 + 含量的变化。结果 PROG预防组和PROG预防并治疗组均能明显降低缺血后脑组织H2 O、Na+ 和Ca2 + 含量的作用 ,PROG治疗组也具有降低脑组织H2 O和Na+ 含量的作用。结论 PROG能降低缺血再灌注性脑损伤脑组织H2 O、Na+ 、Ca2 + 的含量 ,减轻脑水肿 ,从而发挥对缺血脑的保护作用。

Abstract:Objective To confirm the protective effect of progesterone (PROG) on the injury of the focal cerebral ischemia-reperfusion(IR) in rats and to discuss primarily its mechanism. Methods All rats were made to be the model of the focal cerebral ischemia-reperfusion and divided into 6 groups randomly: simple IR group,negative control group ,positive control group , PROG pretreatment group,PROG pretreatment plus post-treatment group and PROG post-treatment group.All rats were decapitated after the left middle cerebral artery was blocked for 2 hours and reperfused for 22 hours respectively,and then ,brain H 2O,Na ,K and Ca 2 contents were determined. Results PROG possessed the significant effects on decreasing brain H 2O,Na and Ca 2 contents in PROG pretreatment group and PROG pretreatment plus post-treatment group in rats . PROG posttreatment group also could lower brain H 2O and Na contents in rats. Conclusion PROG could achieve the protective effects on brain injury by ameliorating the contents of H 2O,Na and Ca 2 of IR brain tissue and reducing brain edema in IR rats.
