摘    要:
为了解人乳头瘤病毒(HPV)对免疫Langerhans细胞(LC)的影响及HPV的致癌机理,采用地高辛配基标记的HPV16/18DNA探针和抗S100蛋白抗体,分别用原位分子杂交和免疫组化LSAB法,检测了40例食管鳞状细胞癌中HPVDNA序列及分布,重点观察了HPV感染时局部癌旁上皮内S100蛋白阳性Langerhans细胞的分布、数量及形态改变。结果显示:在有HPV感染时,上皮内LC数量明显少于无感染者。上皮内残留的少数LC的分布及形态也有改变。结果提示:HPV可能通过减少局部的LC,破坏机体局部的免疫监视系统,并与其他致癌因素协同作用,进而导致食管癌的发生。

Abstract:
The interrelation between human papillomavirus (HPV) infection and Langerhans cells (LC) in the carcinogenesis of esophageal carcinoma and the oncogenic mechanism during HPV infection were studied in this study.Digoxigenined HPV DNA probes of HPV 6B/11,HPV 16/18 and anti s 100 protein antibody were used respectively to investigate the HPV infection and LC with in situ hybridization (ISH) and the immunohistochemical LSAB technique in 40 cases of esophageal squamous cell carcinoma (ESCC).The results showed that there were fewer LCs in the HPV positive cases than those in the HPV negative ones,and even there was nearly not any in the HPV infected foci.The morphological and distributive changes of the LCs in the HPV infected ep ithelia adjacent to cancer also happened.Our results indicate that HPV infection may decrease the number of the local LCs,destroy the local immune surveillance system,and cooperate with other carcinogenic factors,in favor of the development of ESCC.
