银杏叶提取物促进帕金森病模型小鼠室管膜下区神经干细胞增殖作用的研究

摘    要:目的:探讨银杏叶提取物促进帕金森病模型小鼠原位神经干细胞增殖分化的作用。方法:C57BL/6小鼠随机分成正常组、模型组和银杏叶提取物治疗组(简称治疗组)。采用MPTP腹腔注射建立帕金森病小鼠模型,利用免疫组化单标和双标技术观察银杏叶提取物对小鼠侧脑室室管膜下区神经干细胞增殖分化的影响。结果:银杏叶提取物治疗7d,侧脑室室管膜下区的PCNA、Nestin阳性细胞较模型组明显增多,并可见大量从背外侧角沿胼胝体排列的PCNA、Nestin阳性细胞;银杏叶提取物治疗28d,PCNA/Nestin双标细胞的数量较模型组明显增多。结论:银杏叶提取物可促进帕金森病模型小鼠脑内神经干细胞的增殖和迁移。

Abstract:Objective :To explore the role of Ginkgo Biloba Extract (GBE)in promoting proliferation and differentiation of neural stem cells (NSCs)in vivo in modeled mice of Parkinson's disease. Methods:C57BL/6 mice were randomly divided into normal group,model group and GBE treatment group (treatment group in short term).i.p MPTP was adopted to establish Parkinson's disease model in mice.Single-labeled immunohistochemistry and double-labeled technique were applied to observe the impacts of GBE on NSCs proliferation and differentiation in subventricular zone (SVZ)on the opposite side. Results: On the 7th day,the numbers of PCNA and Nestin positive cells in GBE treatment group were increased apparently in lateral SVZ compared with those in model group.It was found that a number of PCNA and Nestin positive cells arranged through corpus callosum from lateral dorsal horn in GBE group. On the 28th day,the number of PCNA/Nestin double-labeled cells in GBE treatment group was much more increased than that in model group. Conclusion:GBE promotes proliferation and migratory of NSCs in brain of Parkinson's disease modeled mice.
