补阳还五汤对脑缺血再灌注大鼠皮质神经细胞凋亡的影响

摘    要:目的 研究补阳还五汤对脑缺血再灌注大鼠皮质神经细胞凋亡的影响。方法 将 36只SD大鼠随机分为 3组 :假手术组 (P ,n =12 )、模型对照组 (C ,n =12 )、补阳还五汤组 (B ,n =12 )。线栓法制作大鼠脑缺血再灌注模型 ,脑缺血再灌注 4 8h后TTC染色计算脑梗死体积 ,TUNEL染色检测神经细胞凋亡 ,免疫组化方法检测缺血侧皮质Bcl- 2 /Bax的表达 ,电镜观察线粒体的变化。结果 脑缺血再灌注后缺血侧皮质神经细胞凋亡增多 ,同时Bcl- 2表达减少 ,Bax表达增多 ,Bcl- 2 /Bax比值下降 ,线粒体肿胀且大多破裂。补阳还五汤干预后 ,细胞凋亡减少 ,脑梗死体积减小 ,Bcl- 2 /Bax比值上升 ,线粒体肿胀减轻。结论 补阳还五汤可能通过抑制神经细胞凋亡而减小脑梗死体积 ,其机制可能是通过上调Bcl- 2 /Bax比值 ,保护线粒体 ,防止发生线粒体通透性转变。

Abstract:Objective It is to study the influence of Buyang Hua nwu decoction on b rain cortex apoptosis of cerebral ischemia and reperfusion rats. Methods 36 SD r ats were randomly divided into sham-operation group (P, n=12), control grou p (C, n=12) and Buyang Huanwu decoction group (B,n=12). Cerebral ischem ia and reperf usion rat models were made using the method of thread inserting right middle cer ebral artery occlusion. The infarct volume was measured 48 hours after reperfusi on. At the same time, the apoptosis was detected by TUNEL dyeing. The expression of Bcl-2 and Bax in different groups was detected by means of immunohistochemis try. The change of mitochondrion was observed with electriomicroscopic. Results The apoptosis in ischemic brain cortex was increased and the expre ssion of Bcl-2 was decreased and Bax was increased. The ratio of Bcl-2/Bax was decreased. The swelled mitochondria were lightened. When the model rats were interfered by Buya ng Huanwu decoction, the apoptosis decreased, the brain infarct volume di minished, the ratio of Bcl-2/bax increased, and the swelled mitochondria w as lightened. Conclusion Buyang Huanwu decoction can diminish brain infarct vol ume possibly b y inhibiting apoptosis. The mechanism may be increasing the ratio of Bcl-2/bax, protecting the mitochondrion and preventing the mitochondrion permeability trans ition.
