人大肠癌中Ⅰ型膜型金属蛋白酶mRNA的表达

目的:探讨Ⅰ型膜型金属蛋白酶(membrane-type matrix metalloproteinase 1,MT1-MMP)mRNA的表达与人类大肠癌的关系.方法:用逆转录聚合酶链式反应(RT-PCR)对38例大肠癌中MT1-MMP mRNA的表达进行检测.结果:86.8%(33/38)的大肠癌中MT1-MMP mRNA的表达高于相应正常黏膜(P＜0.01),癌/ 膜的中位数为1.63(Min=0.70,Max=11.00),其表达水平与大肠癌的浸润深度呈正相关(P＜0.05).结论:MT1-MMP高表达可能在大肠癌对周围组织的浸润过程中具有重要作用,可以作为反映大肠癌侵袭能力的生物学标志物.

Expression of membrane-type matrix metalloproteinase 1 ( MT1-MMP ) mRNA in human colorectal carcinomas

Objective: To investigate the correlation of MT1 MMP mRNA expression with the progression of colorectal carcinomas. Methods: RT PCR technique was used to measure the level of MT1 MMP mRNA in 38 cases of colorectal carcinoma. Results: In 86.8% (33/38) of the cases, the expression of MT1 MMP mRNA in carcinomas was higher than that in paired adjacent normal mucosas ( P <0.01), and the median value of the tumor normal ratio was 1.63 (Min=0.70, Max=11.00). The tumor normal ratio of MT1 MMP mRNA was positively correlated with the depth of tumor invasion( P <0.05). Conclusion: The over expression of MT1 MMP may play an important role in the invasion of colorectal carcinoma, and may be used as a biologic marker in predicting the aggressiveness of colorectal cancers.
