百合多糖联合大蒜素对肝癌H22荷瘤小鼠肿瘤生长及血清细胞因子水平的影响

摘    要:
目的:探讨百合多糖联合大蒜素对肝癌H22荷瘤小鼠肿瘤生长及血清细胞因子水平的影响。方法:雄性 BALB/c小鼠腋下注射肝癌H22细胞计数瘤液建立荷瘤模型,分为百合多糖组(100 mg/kg)、大蒜素组(40 mg/kg)、百合多糖低剂量+大蒜素组(50 mg/kg+40 mg/kg)、百合多糖中剂量+大蒜素组(100 mg/kg+40 mg/kg)、百合多糖高剂量+大蒜素组(150 mg/kg+40 mg/kg)、正常组、模型组和环磷酰胺组(30 mg/kg)。处理7 d后,观察小鼠的肿瘤重量,计算抑瘤率;胸腺、脾脏称重,计算胸腺指数和脾脏指数;ELISA法检测血清免疫相关细胞因子IL-18、TNF-α、IFN-γ、IL-2、IL-6水平及促血管生长因子VEGF水平。结果:大蒜素组、百合多糖中剂量+大蒜素组、百合多糖高剂量+大蒜素组抑瘤率分别为27.27%、34.68%和30.64%。百合多糖中剂量+大蒜素组和百合多糖高剂量+大蒜素组胸腺指数明显高于模型组(P<0.01)。百合多糖高剂量+大蒜素组较模型组明显升高IL-18水平(P<0.01)。百合多糖中剂量+大蒜素组TNF-α水平高于模型组(P<0.05)。百合多糖中剂量+大蒜素组较模型组明显升高IL-2水平(P<0.01)。百合多糖高剂量+大蒜素组较模型组降低VEGF水平(P<0.05)。结论:百合多糖联合大蒜素能抑制肝癌H22荷瘤小鼠肿瘤的生长,提高荷瘤小鼠的胸腺指数,并促进小鼠分泌免疫相关细胞因子IL-18、TNF-α、IL-2,减少分泌VEGF。

Abstract:
Objective:To investigate the effect of lily polysaccharide combined with allicin on tumor growth and serum cytokines level in mice bearing liver cancer H22.Methods:Male BALB/c mice were injected with liver cancer H22 count tumor solution under the armpit to establish a tumor-bearing model.They were divided into lily polysaccharide group(100 mg/kg),allicin group(40 mg/kg),low-dose lily polysaccharide+allicin group(50 mg/kg+40 mg/kg),medium-dose lily polysaccharide+allicin group(100 mg/kg+40 mg/kg),high-dose lily polysaccharide+allicin group(150 mg/kg+40 mg/kg),normal group,model group and cyclophosphate amide group(30 mg/kg).After 7 days of treatment,the tumor weight of nude mice was observed,and the tumor inhibition rate was calculated.The thymuses and spleens were weighted.The thymus index and spleen index were calculated.ELISA method was used to detect serum cytokines IL-18,TNF-α,IFN-γ,IL-2,IL-6 level and VEGF level.Results:The tumor inhibition rates of allicin group,medium-dose lily polysaccharide+allicin group,high-dose lily polysaccharide+allicin group were 27.27%,34.68% and 30.64%,respectively.The thymus index of the medium-dose lily polysaccharide+allicin group and the high-dose lily polysaccharide+allicin group was significantly higher than that of the model group(P<0.01).Compared with the model group,the high-dose lily polysaccharide+allicin group significantly increased the level of IL-18(P<0.01).The level of TNF-α in the medium-dose lily polysaccharide+allicin group was higher than that in the model group(P<0.05).The level of IL-2 in the medium-dose lily polysaccharide+allicin group was significantly higher than that in the model group(P<0.01).The high-dose lily polysaccharide+allicin group lowered the level of VEGF than the model group(P<0.05).Conclusion:Lily polysaccharide combined with allicin could inhibit tumor growth in mice bearing liver cancer H22,increase the thymus index of tumor-bearing mice,promote the secretion of cytokines IL-18,TNF-α and IL-2 and reduce the secretion of VEGF in mice.
